Practice and exploration of integrating ideological and political education into course teaching of Hydraulics

Since the 18th National Congress of the Communist Party of China, General Secretary Xi Jinping emphasized that moral education should run through the whole process of education, and that all kinds of courses should go together with ideological and political courses to form synergistic effect. Taking Hydraulics as an example, this paper made a preliminary exploration into the ideological and political teaching of specialized courses. The ideological and political elements contained in Hydraulics were deeply explored, and the ideological and political education was integrated into the classroom teaching of hydraulics from the aspects of China’s achievements in Hydraulics, scientists’ deeds, typical water conservancy projects, life practice, engineering ethics and classical culture. In a silent way, the students were taught the socialist core values, academic aspirations and professional ethics. The teaching mode and carrier approach of hydraulics course were innovated and improved the college student’s acquisition sense of ideological and political education.

Study on the sensitivity of Ichthyophthirius multifiliis to three chemical drugs

In this study, Ichthyophthirius multifiliis isolated from Cyprinus carpio was in vitro monoclonal culture, using mixture of copper sulfate and ferrous sulfate(5:2), aqueous solution of potassium permanganate and formalin to explore the mortality to Ichthyophthirius multifiliis. The results showed that the concentration of the three drugs is positively correlated with the mortality of Ichthyophthirius multifiliis, and 5.6 mg/L of copper sulfate and ferrous sulfate mixture, 8 mg/L potassium permanganate aqueous solution and 120 mg/L formalin can kill all of the Ichthyophthirius multifiliis. The sensitivity of the Ichthyophthirius multifiliis to these three chemical drugs: Copper sulfate and Ferrous sulfate mixture>Potassium permanganate aqueous solution>Formalin. The research results provide corresponding theoretical data and experimental basis for the treatment and prevention of Ichthyophthirius multifiliis disease to a certain extent.

Effect of chemical fertilizer reduction on the availability of microelement in facilities vegetable soil

In order to study whether reducing the amount of chemical fertilizers will affect the availability of microelement in the facilities vegetable soil, the change of available Fe, Mn and Cu contents in the soil of the facilities vegetable of Shuigaozhuang and Xiaoshawo village of Tianjin were studied by reducing fertilizer application on the basis of applying organic fertilizer, so as to provide reference for rational fertilization of facilities vegetable. The results indicated that the available Fe content was the highest in the 10% chemical fertilizer reduction treatments, and was the lowest in the 25% chemical fertilizer reduction treatments both in the soil of the winter melon shed in Shuigaozhuang and the celery shed in Xiaoshawo; the available Fe content was the highest in the 25% chemical fertilizer reduction treatment in the soil of the tomato shed in Xiaoshawo; the winter melon shed in Shuigaozhuang village and the celery shed in Xiaoshawo had the highest available Mn content when the chemical fertilizer was reduced by 25%, while the tomato shed in Xiaoshawo when reduced by 35%. The available Cu content in the soil of the three vegetable sheds was the lowest with no chemical fertilizer reduction.

As a new teaching management system, credit system plays an important role in promoting China’s educational reform and personnel training. The reform of the credit system in our country shows the characteristics of paying attention to the development of students’ personality, adopting the flexible educational system, establishing the tutorial system, carrying out the credit system charging and emphasizing the management by objectives. The reform of the credit teaching management system is an important measure to promote the quality of undergraduate education in colleges and universities. The implementation of the credit system is conducive to improving the motivation in the reform of the teaching management system of colleges and universities, which is the breakthrough to deepen the reform of the teaching management system of colleges and universities in China. At the same time, the implementation of the credit system also needs the reform of university teaching management system to ensure the sound operation of the credit system. To reform the credit system teaching management mode, it is necessary to clarify the key factors of the credit system management reform, change the education concept, sort out the working relationship and the system mechanism, innovate the management system, and formulate the institutional system and the corresponding talent training program that adapt to the credit system education mode.

The COVID-19 pandemic has had a great impact on China and the world. In order to fight against it, China has adopted a series of decisive and effective anti-pandemic measures with outstanding results. The leadership of the Communist Party of China, the system of concentrating on doing great things, and the people-centered development concept fully demonstrate China’s unique political and institutional advantages; the core values of socialism play an important leading role in the pandemic prevention and control; the principle of collectivism is fully demonstrated. The ideological and political course can be well constructed with the “anti-pandemic” stories in China. The facts can reveal some western media’s distorted reports on the achievements of China’s “anti-pandemic”, enhance the ideological and political course teachers’ sensitivity and discerning power in keeping the ideological “responsible field”, and cultivate college students’ responsibility and sense of mission to be “responsible for the great task of national rejuvenation”.

The course of Introduction to Human Resource Management should convey correct world outlook, life outlook and life values through “ideological and political education”, and focus on discussing the teaching reform of ideological and political education from four interlocking perspectives: detailed decomposition of ideological and political elements of curriculum, organic integration into teaching content, optimization of teaching means and improvement of curriculum assessment design. The implementation of ideological and political education should take implicit infiltration as the main way, and establish the content of ideological and political education from four aspects: political orientation, core values, professional ethics and learning ethics. It should integrate professional knowledge into ideological and political elements from three dimensions of “managing others” “realizing self-value” and “cultural confidence”, comprehensively apply case teaching, experiential teaching, new technology and other teaching means, and organically integrate routine evaluation and institutional evaluation, so as to promote the compatibility of professional knowledge education and ideological and political education, and help training talents with both ability and morality.

Quality difference analysis of 5 pear varieties on mild saline and alkaline land

In order to screen out the varieties of pears with good flavor to adjust the variety structure of Tianjin pear fruit. The 5 varieties of pear fruits on mild saline-alkali land were rated as research materials. The quality indexes of the pear fruits were analyzed ,such as taste, flavors, single fruit weight, soluble sugar, titratable acid, water content and stone cells, etc. The results showed that the sensory quality of the ‘Huangguan’ pear was good, and the ‘Jin Xiangmi’ was poor. The soluble protein and soluble sugar of the ‘Huangguan’ were the highest(0.62 mg/g, 10.88 mg/100g), but its hardness and water content were the lowest(1.76 kg/cm 2, 81.94%); The water content and soluble solid of ‘Akitsuki’ were the highest(85.57%, 17.98%), but its soluble sugar, Vc and stone cells were the lowest(10.38 mg/100g, 1.19 mg/100g, 0.09%); The hardness and Vc content of ‘Jin Xiangmi’ were the largest(4.61 kg/cm 2, 4.89 mg/100g). The stone cells content of ‘Hong Xiangsu’ was the highest(0.35%), but its single fruit weight and titratable acid content were the smallest(239.59 g, 0.06%). The single fruit weight and titratable acid content of ‘Zao Suhong’ were largest(441.17 g, 0.16%), but its soluble solids and soluble protein were the smallest(11.98%, 0.27 mg/g). According to comprehensive analysis of the index membership function, the comprehensive quality of ‘Akitsuki’ was the best, followed by the ‘Huangguan’, and ‘Jin Xiangmi’, ‘Hong Xiangsu’ and ‘Zao Suhong’ were poor, indicating that ‘Akitsuki’ pear and ‘Huangguan’ pear could be used as the main planting varieties of Tianjin. However, ‘Huangguan’ in view of the difference on the color and smoothness of the peel should be main variety in Tianjin.

Response of interferon α gene to Aeromonas veronii infection in Clarias gariepinus

In this paper, real-time quantitative PCR technique was used to assess the expression profiles of interferon alpha(IFNα)gene in spleen, kidney, head kidney, gill, intestine and skin of Clarias gariepinus following an intraperitoneal injection of Aeromonas veroni. The results showed that the IFNα gene was expressed in all tissues of the healthy(control)individuals, and the highest expression was found in spleen, which was significantly higher than those in other tissues except kidney and skin. After A. veronii challenge during 3-96 h, the temporal expressions profiles of IFNα gene in the six tissues were similar, namely, transcripts were up-regulated firstly and then recovered back to the baseline, but the expression peaked at different time points. Furthermore, the expression levels of IFNα gene were different among the tissues at the same time points of post infection.

In recent years, biocontrol of plant disease has been widely concerned in the world. In order to enrich the resources of antagonistic microorganisms against plant diseases, the Chinese herbal medicine Plantago asiatica was used as experimental material to isolate the endophytic antagonistic bacteria, with Valsa mali, Alternaria solani, Fusarium graminearum, Sphaerotheca fuliginea and Botrytis cinerea used in this study. 20 strains of endophytic bacteria were tested by confrontation method. The results showed that 5 strains of endophytic bacteria showed strong inhibitory effect on the tested pathogens, especially R-6 strains. The protective and therapeutic effects of fermentation products of R-6 on strawberry gray mold and cucumber powdery mildew were 56.9%, 50.5% and 49.0%, 37.3%, respectively. According to its morphological characteristics, physiological and biochemical characteristics and 16S rRNA sequence, the R-6 strain may belong to Bacillus velezensis.

Waste oil is one of the food safety problem which people pay more attentiorto the complexity and non-quantification of waste oil brings great difficulties to its detection. This article introduced the physical and chemical properties of the waste oil, introduced the detection technology based on different physical or chemical properties of the waste oil, and then compared and summarized the advantages and disadvantages of several typical waste oil detection technologies. The spectrum technology has high sensitivity and accuracy, so the spectrum detection technology was mainly introduced from fluorescence spectrum, Raman spectrum, Fourier transform infrared spectrum and terahertz spectrum. By comparing various detection technologies, it concluded that the fluorescence spectrum will have great potential in the detection of waste oil in the future.

Graduation thesis is the last indispensable practical link in the process of undergraduate education. In view of the problems in the current graduation thesis, the reasons for the unstable quality of graduation thesis were analyzed, combined with the requirements of hydrology and water resources engineering education professional certification, and a “staged” graduation thesis assessment system was constructed. The whole graduation thesis process was divided into three stages: the beginning, mid-term and defense, and the corresponding graduation requirements index points for each stage, assessment and evaluation item index system, index weights, assessment and scoring method were clarified. The graduation thesis assessment system should be strengthened strictly to improve the overall quality and ability of graduates.

In Escherichia coli, signal recognition particle(SRP)recognizes and binds signal peptides being translated by ribosome to guide the new peptide chain to the cell membrane. In this experiment, localization of E.coli membrance protein in HDB51 and MG1655 was studied by Western blot based on biotinylation for membrane proteins targeting. By comparing the presence or absence of SRP in different E.coli, the substrate protein FtsQ that depended on SRP in the strain E. coli HDB51 was localized. The location of FtsQ was significantly affected without SRP. This verifies that SRP helps membrane protein localization. It can provide references for the study of localizing E. coli membrane proteins dependent on SRP.

Cloning and construction of prokaryotic expression plasmid of truncated SRS29C gene of Toxoplasma gondii

In order to construct the prokaryotic expression plasmid of Toxoplasma gondii SRS29C gene, the signal peptide of SRS29C gene was predicted by using SignalP-5.0 software, the sequence of the signal peptide of SRS29C gene was removed, the truncated gene of SRS29C was amplified and cloned, and then the prokaryotic expression plasmids were constructed. The results showed that the first 51 amino acids of SRS29C were the signal peptide region, so by cutting off the signal peptide, the 966 bp SRS29C truncated gene fragment (SRS29Ccut) was successfully obtained, and the recombinant plasmids pET-28a-SRS29Ccut and pET-32a-SRS29Ccut were successfully constructed. This study may lay a foundation for further study on the biological function and immune characteristics of the protein of Toxoplasma gondii SRS29C.

Biogas production rate in the bench-scale fermentation reactor is commonly low, and it is difficult to obtain the precise data of biogas production. This study designed a biogas flow meter that is suitable for bench-scale fermentation reactor. During the operation of the flow meter, the continuously produced biogas is discretized and then measured, and the biogas volume in each measurement batch is determined by the pressure change in the biogas collection chamber with the constant volume. The control system of the flow meter takes the single chip microcomputer as the core part. Based on the predetermined pressure parameters, the automatic switch of the situation between “receiving biogas” and “emitting biogas” in the biogas collection chamber could be achieved. The data of the key parameters that is automatically collected and displayed in the control system is used for the calculation of biogas flow.

Furthermore, a list of all the known species and identification key of Encyrtidae in Tianjin Agricultural University were given, and other detailed information about hosts and geographic distributions were also provided.

Induction of loose embryogenic callus from Anthurium andraeanum

In order to obtain the loose embryogenic callus of Anthurium andraeanum, different explants, different hormone treatment combinations and MS medium inorganic salt multiple were used to induce the embryogenic callus of Anthurium andraeanum. The results showed that the leaves of Anthurium andraeanum were the best explants for callus induction, and the loose callus could be induced on the medium of 1/2MS+2.0 mg/L 2,4-D+0.2 mg/L NAA, and transferred to MS+1.0 mg/L BA+1.0 mg/L 2,4-D medium, subcultured once every 15 days, and subcultured 2-3 times, and then the loose embryogenic callus of Anthurium andraeanum could be obtained and regenerated directly on MS medium. The induction system of loose embryogenic callus of Anthurium andraeanum was established in this experiment, which can lay a foundation for the study of tissue culture and in vitro mutagenesis of Anthurium andraeanum.

Induction of polyploid Robinia pseudoacacia L. and analysis of anatomical structure of leaves

Tetraploid Robinia pseudoacacia L. has been widely planted because of its strong adaptability and large biomass. In this study, the seedlings of R. pseudoacacia were treated with 5 different concentrations colchicine to induce tetraploid, and the changes of the anatomical structure of the tetraploid leaves after induction were studied. It was found that 0.05% colchicine could achieve better induction effect in one day. The thickness of the upper epidermis, the palisade tissue, sponge tissue and leaf thickness of the tetraploid R. pseudoacacia leaves were significantly higher than those of diploid, and the cell tense ratio of tetraploid ones were significantly improved, which indicated that R. pseudoacacia had strong resistance to stress after tetraploidization. The results can provide reliable technical support for the polyploid breeding of R. pseudoacacia, provide reference and theoretical support for the ploidy breeding of R. pseudoacacia, and provide morphological and anatomical basis for explaining the formation principle of the resistance of tetraploid R. pseudoacacia.

One would be responsible to do something, can be responsible to do something, must be responsible to fulfill something and will be responsible never to do any corruption. One should set a good example by personal taking part in and being steadfast to the work. Lastly, oneshould politically self-disciplined by distinguishing multiple corruptions. All of us must build the rules, keep the laws, refuse corruption, and defend integrity and self-discipline.

Study on the preparation methods of homogenate chlorella cells

With Chlorella. vulgaris as the test organism, four homogenizate methods ,liquid nitrogen method, grinding method, mechanical method, and repeated freeze-thaw method were studied. The homogenation with the above methods was used to determined the SOD activity and GSH content, which aimed to select a method with high crushing efficiency, convenience, and accurate determination of physiological and biochemical indicators. The results showed that the crushing rates of the four methods were 81.43%, 100%, 92.41%, 56.54%, and the SOD activity were 47.06, 48.58, 0, 25.05 U/mgprot. There was no significant difference between liquid nitrogen method and grinding method( p>0.05); GSH content were 34.42, 24.52, 87.37, 38.75 mgGSH/gprot; There was no significant difference apart of mechanical method( p>0.05). The above results indicated that the grinding method and liquid nitrogen method have a high crushing rate, and accurate measurement of the biochemical indicators. However, grinding method consumed too much labor and cannot process a large number of samples, so the liquid nitrogen method was considered to be the best solution for preparing microalgal cell homogenation.

Comparative study on cultivation techniques of dry direct seeding rice and conventional transplanting rice

With the restriction of water, soil, environment and other factors, the dry direct seeding cultivation technology of rice is developing day by day. By comparing the differences of cultivation methods, water and fertilizer management, yield and quality, and soil properties between dry direct seeding rice and conventional transplanting rice, the results showed that dry direct seeding rice had fast growth process, high water production efficiency, and that nitrogen use efficiency could be improved by appropriately reducing nitrogen in the early stage. The difference of yield between dry direct seeding and conventional rice was not significant, but protein content of dry direct seeding rice was increased, and its soil aeration was good. It had advantage on structure improvement and fertility maintenance, water saving, labor saving, time saving and soil environment improvement. At the same time, it was pointed out that there were some problems such as lack of effective control of diseases, pests and weeds, lodging easily, and unstable yield and quality. Finally, the development prospects of dry direct seeding and conventional cultivation techniques were analyzed from different perspectives. It was proposed that in the production areas with prominent constraints of soil and water environment, with the promotion of pest control technology and variety breeding, dry direct seeding technology will achieve the great potential of sustainable production and become an effective way to ensure food security in China.

In this study, the F 2 generation population constructed by the cross between the grain Sorghum bicolor ‘Xinliang 52’ and Sorghum sudanense ‘Meiyin-48’ was used as the test material. Five agronomic characteristics of the F 2 generation including leaf numbers, stem numbers, plant height, stem diameter and wax powder were measured, and the optimal genetic model and heritability of each trait were obtained by principal-multiple gene analysis. The results showed that the most suitable model for the number of leaves and stem diameter was Model A_0. There was no major gene inheritance, but the quantitative trait inheritance controlled by micro-effect and multi-gene. The optimal model of stem numbers and plant height was Model A_4, a negative full-dominant model controlled by a pair of major genes, and the major gene heritability were 91.13% and 66.14%. The optimal model of wax powder was Model B_1, which belonged to the additive-dominant-epistatic mixed genetic model, controlled by two pairs of major genes with the heritability of the major gene of 54.03%.

Antibacterial activity of Earthworm homogenate supernatant on common harmful bacteria in pig farm

With the advent of the era of feed without antibiotics, the research and development of feed without antibiotics or alternative antibiotics has become a hot spot of current research. Earthworms are high–quality protein feed, and Chinese medicine believes that earthworms have a number of therapeutic effects. The aim of this study was to determine the antibacterial activity of earthworm homogenate on Escherichia coli, Staphylococcus aureus, Streptococcus and Haemophilus parasuis from pig farm, with the antibacterial activity of earthworm homogenate treatment solution studied by Kirby-Bauer test(K-B)and ultraviolet spectrophotometry. The study showed that the supernatant of the earthworm homogenate had antibacterial effects on Escherichia coli, Staphylococcus aureus, Streptococcus and Haemophilus parasuis. The highest concentration of earthworm homogenate supernatant with a ratio of earthworm to water of 2:1 has antibacterial activity on Escherichia coli. The supernatant of earthworm homogenate had obvious and broad–spectrum antibacterial activity effect on Staphylococcus aureus. The ratio of earthworm to water 4:5 had obvious antibacterial activity on Streptococcus and Haemophilus parasuis. The spectrophotometric test showed that two different concentrations of supernatant of earthworm homogenate had different degrees of bacteriostatic effect on Escherichia coli, Staphylococcus aureus and Streptococcus. Earthworm homogenate 2 was more effective. 24 hours later, the antibacterial advantage of earthworm homogenate supernatant disappeared completely. This study showed that the supernatant of earthworm homogenate has certain bacteriostasis to common harmful bacteria in pig farm, which can provide theoretical basis for the development of new feedstuffs related to earthworms.

Isolation, identification and drug resistance of canine Staphylococcus pseudointermediate

An 11-year-old golden-haired dog with pyoderma was admitted to a pet hospital in Tianjin. The secretions of eyes, nose and skin were collected for bacterial isolation and culture. The results of culture characteristics, staining microscopy, biochemical test, plasma coagulase test, drug susceptibility test and 16S rDNA sequence analysis showed that the isolate TJ-1 decomposed fructose, lactose, mushroom sugar, sucrose, mannose, maltose, mannitol and nitrate reduction were negative, while plasma coagulase test was positive. The 16S rDNA sequence was amplified by PCR and sequenced. The phylogenetic tree analysis showed that the isolate was clustered with Staphylococcus pseudintermedius and TJ-1 strain was identified as S. pseudintermedius. Drug susceptibility test showed that the bacteria were susceptible to oxacillin, ceftazidime, cefoperazone, cefalexin, doxycycline, cefazolin, minocycline, cefradine, cefuroxime and amikacin, but resistant to erythromycin, penicillin, ampicillin and carboxybenicillin.

According to the statistical data of the questionnaire on students in four universities in Tianjin, the mainstream of the college students’ political belief is positive while someone has belief crisis. Most students believe in Marxism, but their understanding of Marxist theory is not deep enough, and the theoretical basis of Marxism is not firm enough. College students highly identify with socialism with Chinese characteristics, and they are full of confidence in the ruling ability of the Communist Party of China. They have a strong desire to join the Party, but their motivation is utilitarian. Ideological and political courses are the main way of belief education in universities, but the educational effect is not ideal enough. In view of the actual situation of college students' political belief, universities should strengthen the education of political belief, do a good job in the ideological and political teaching, and make good use of the class which is a main channel of teaching. The quality of teachers of ideological and political courses should be improved so that people with belief can speak their beliefs. Attention should be paid to the cultivation of practical education and the function of cultivation in social practice should be fully played. The cultivation through campus culture and integrating the belief education in community activities should be strengthened. The construction of network should be improved to promote the organic combination of new mass media and belief education, and enhance the radiation of belief education.

To explore the cause of death of Epinephelus coioides, a bacterial strain was isolated from the liver of diseased Epinephelus coioides and named as SBY-G. This bacterial strain was preliminarily determined as Vibrio harveyi according to its morphological, physiological and biochemical characteristics. The sequence analysis of the 16S rDNA and gapA genes respectively showed SBY-G had the highest similarity with V. harveyi(KU364055.1)and(LC370081.1), and the phylogenic trees showed that it was clustered with V. harveyi. The results of antibiotic susceptibility analysis showed that SBY-G was highly sensitive to amoxicillin, cefixime, cefamandole, ceftriaxone, norfloxacin and other fifteen antibiotics, moderate sensitive to cefotaxime, gentamicin, kanamycin and other six antibiotics, while resistant to ampicillin, oxacillin, cefuroxime, ceftazidime, streptomycin, neomycin and other eighteen antibiotics. Artificial infection experiments showed that the median lethal dosage(LD 50)of the SBY-G strain on zebra fish was calculated as 1×10 5 cfu/g. The results of this study provide a scientific reference for the prevention, treatment and diagnosis of fish diseases caused by V. harveyi.

In the process of planting green plants, cultivation matrixes have important effects on plant growth. Therefore, it is very important to develop a substrate with strong fertilizer supply capacity, pH and EC suitable for the growth of green plants. In this experiment, with Quamoclit pennata as a green plant, five different cultivation matrix formulations using Fluvo-aquic soil, peat and vermiculite were made to study the changes of available nutrients in the growth of Q. pennata in different cultivation matrixes, and the growth conditions and seedling indexes of Q. pennata in different cultivation matrixes were analyzed. The results showed that the cultivation matrix of “Fluvo-aquic soil 50%+peat 30%+vermiculite 20%” and “Fluvo-aquic soil 40%+ peat 60%” had strong and stable ability to supply nitrogen, phosphorus and potassium. The pH and EC were also suitable for the growth of Q. pennata, and the seedling height, stem diameter, dry weight and strong seedling index were all higher. Thus, both of the above two substrates can be used as the cultivation matrixes of Q. pennata.

Effects of two kinds of diets and feeding frequency on growth, digestion and immune response of Portunus trituberculatus

A 28 d feeding trial was conducted to study the effects of different diets and feeding frequency on the growth, digestion and immune indexes of Portunus trituberculatus. A two-factor experimental design was adopted with Aloididae aloidis and black soldier fly larvae as baits and the feeding frequency set as twice/d and 3 times/d, respectively. 36 crabs with an initial weight of (75.8±16.8) g were randomly divided into 4 groups with 3 replicates in each group and 3 crabs in each replicate. The results showed that feeding frequency and diet type did not significantly affect the weight gain rate and specific growth rate of Portunus trituberculatus. Feeding black soldier fly significantly increased the hepatopancreas index( P<0.05). The protease and cellulase activity of crab were significantly influenced by feeding frequency and diet type( P<0.05), and the feeding frequency also affected the amylase activity( P<0.05). The feeding frequency and diet type did not influence the acid phosphatase( ACP)and total superoxide dismutase( T-SOD)activity of crab, but showed a significant effect on the activity of alkaline phosphatase( AKP)( P<0.05). Also, the diet type showed a significant effect on the Catalase( CAT)activity, which was higher in crab fed with black soldier fly than that fed with aloididae aloidis( P<0.05). In conclusion, feeding frequency did not significantly affect the growth performance of Portunus trituberculatus. There was no significant effect on the growth performance of Portunus trituberculatus no matter when fed with black soldier fly larvae or Aloididae aloidis. However, the non-specific immunity of crab fed with black soldier fly larvae improved. Generally, it was possible in practice to feed Portunus trituberculatus with Black soldier fly larvae instead of Aloididae aloidis.

An immunosensor fabricated by immobilizing Brucella antibody onto the surface of gold nanoparticles-modified screen-printed carbon electrodes(GNPs-SPCEs)was proposed for the rapid detection of Brucella. The gold nanoparticle carbon electrode was used as the working electrode, and the Brucella antibody was modified on the surface of the working electrode to construct the electrochemical three electrode system. The reaction process was characterized by cyclic voltammetry(CV). The change value of oxidation peak current(Δ IPA)was used as the current response signal, and PBS buffer was used as blank control. It was found that when the antigen of Brucella combined with antibody, the oxidation peak current decreased, while the current value of sterile PBS buffer almost did not change. When Escherichia coli and Staphylococcus aureus were used to replace Brucella, the response signal of interfering bacteria was similar to that of blank solution. Therefore, the immunosensor has good specificity for Brucella.

The new agricultural format is an important way to transform and upgrade modern urban agriculture in Tianjin. At present, a variety of new types of agricultural formats represented by leisure agriculture in Tianjin have made great progress, and their formation paths mainly include: horizontal expansion of industrial chain based on the consumption demand, vertical extension of industrial chain based on increasing added value, and penetration in agriculture based on the integration of modern technology. However, the development of new agricultural formats in Tianjin is facing many difficulties, and it mainly includes: the imperfection of land policies, the urgent need for using technological innovation to promote the integration of three industries, and the lag of agricultural multi-functional development and brand building. In order to break through the above difficulties, land policy should be improved to guarantee the standard of basic farmland, scientific innovation should be strengthened to promote industrial integration, talents should be attracted to adopt new marketing methods and multiple functions of agriculture should be explored to make it more dynamic.

Miamiensis avidus is a common marine pathogentic ciliate which can multiply quickly in eutrophic waters, causing fish disease and great loss to fishery resources and aquaculture. There are no drugs to prevent fish from being infected until now. In recent years, Sodium chloride, Mercury chloride, Trichlorfon, Cupric sulfate have been commonly used in aquaculture, but there was no report on the toxicity of four drugs to animals low on the aquatic food chain. In this study, the toxicity of four drugs to M. avidus was investigated . M. avidus was cultured to a density of 2 500 ind/mL, at 25℃ and a light intensity of 1 000 lx. Based on the result of pre-test , the effect of four drugs(sodium chloride: 0, 200, 400, 600, 800, 1 000 mg/L; Mercury chloride: changes in population density at 0, 0.002, 0.004, 0.006, 0.008, 0.010 mg/L; Trichlorfon: 0, 100, 200, 300, 400, 500 mg/L; Cupric sulfate: 0, 0.010, 0.015, 0.020, 0.025, 0.030 mg/L)was studied. The tests showed that four drugs all inhibited the population growth of M. avidus. Respectively, the sodium chloride 800 mg/L, Trichlorfon 500 mg/L, Cupric sulfate 0.03 mg/L, Mercury chloride 0.01 mg/L, the inhibition of M. avidus population growth is the most obvious.

Comparison of three corrosion methods for cast specimens of tracheobronchial duct in rabbits

To find out an effective and simple method to make the casting specimen of rabbit’s pulmonary trachea and bronchial tree, so as to provide a real and rigorous anatomical basis for veterinary related teaching, scientific research and clinical practice, three corrosion methods were used in this research. Self-solidifying denture powder and denture water were used to prepare fillers in proportion, and then perfusion from the throat into the lungs of rabbits. After the filler solidified, Sodium hydroxide solution, sulfuric acid solution and hydrochloric acid solution were used to corrode the lung tissues. The results showed that all of the three corrosion methods could get good cast specimen. The specimen corroded by sodium hydroxide solution only needed to be immersed in the same solution again; The specimens corroded by sulfuric acid solution were black and needed to be bleached again; The specimens corroded by hydrochloric acid solution needed to be immersed in alkaline solution to remove the residual lipids. After filling the lung with self-coagulating dental powder and dental bracket water at the ratio of 1.0∶1.5, 50% sodium hydroxide solution were used to corrode the lung tissue. The intact, moderately elastic, clean and smooth respiratory cast specimens could be obtained, and bronchial trees could be clearly seen. Sodium hydroxide solution did not need to be equipped with other solutions, and the substance was not volatile. It is the most convenient and safe method among the three corrosion methods.

Comparison of fruit quality of Dongzao jujube from different variety

In order to compare the quality of jujube sold on Tianjin market from different variety, the study selectcd three varieties of Dongzao: Huanghua jujube, Zhanhua jujube and the second generation of Zhanhua jujube and compared the differences of the external quality(single fruit weight, fruit shape index, hardness)and internal quality(water content, soluble solids, soluble sugar, titratable acid, vitamin c). The results showed that the single fruit weight(28.44 g), hardness(20.92 kg/cm 2), soluble solid content(16.73%), soluble sugar content(28.47%), titratable acid content(0.64%)and Vc content(429.57 mg/100g)of the second generation of Zhanhua jujube were the highest. The fruit shape index(1.04), water content(83.05%)and sugar-acid ratio (89.69)of Zhanhua jujube were the highest. The fruit quality of jujube was in the order of the second generation of Zhanhua jujube, Zhanhua jujube, and Huanghua jujube according to a membership function analysis.

The purpose of this paper is to investigate the effects of different atrazine and thifensulfuron on the population dynamics of Euplotes vannus, Uronema marinum and Pseudokerono psiscarnea by calculating the population density, growth rate and generation time of Euplotes vannus, Uronema marinum and Pseudokerono psiscarnea. The results showed that the atrazine and thifensulfuron to Euplotes vannus concentration of 6 mg/L, the maximum population density was higher than other experimental groups. There were differences in population growth rate and generation time for each concentration of Euplotes vannus. The atrazine and thifensulfuron to Uronema marinum concentration of 6.5 mg/L, the population growth was obviously inhibited. The population growth rate and generation time of Uronema marinum at different concentrations were quite different. Atrazine to Pseudokeronopsis carnea concentration of 6.5 mg/L and thifensulfuron to Pseudokerono psiscarnea concentration of 7.5 mg/L, the population growth was obviously inhibited. The population growth rate and generation time of Pseudokerono psiscarnea at each concentration were significantly different.

Study on the clarification technology and aroma analysis of persimmon wine

In order to reduce the turbidity of the persimmon wine and improve the wine quality, natural clarification, membrane filtration clarification, centrifugal clarification and clarification of clarifying agents were used. The results showed that the clarification effect of clarifying agents was the best. The light transmittance was the highest when the chitosan added amount was 0.04 g/L, reaching 96.8%. The electronic nose was used to analyze the aroma components. The column temperature was 40 ℃, the injection port temperature was 200 ℃, the injection port pressure was 25.0 kPa, and the injection port flow rate was 10.0 mL/min. Under these conditions, there was little difference in volatile aroma components in wine between clarification of chitosan and naturally clear persimmon, and the clarification effect is ideal when chitosan is used as a clarifying agent.

In order to research the salt tolerance of walnut rootstocks, so as to provide scientific basis for screening salt-tolerant walnut rootstocks, the seedlings of Juglans microcarpa were treated under salt stress with NaCl and the growth situation of seedlings were observed. The physiological indexes of Juglans microcarpa seedlings were also measured in different days after treatment. The results showed that the walnut leaves were wilted and the leaves became yellow. The leaves become dark until the plants die under high-concentration and long-term salt stress. With the increase of salt concentration and stress time, the activity of POD and SOD increased first and then decreased, the free proline accumulated, and the change trend of soluble protein content was increasing first and then decreasing. It was found that the seedlings of Juglans microcarpa showed tolerance of salt stress with 100 mmol/L NaCl.

Based on the concept of Production-Living-Ecology Space, the Characteristic Towns of Tianjin are classified into three types: production type, life type and ecological type. The integrated development of the Production-Living-Ecology Space still faces the dilemma of imperfect industrial system, insufficient attraction of living space and great threat to ecological space. Based on the CAS theory, the integrated development mechanism of Production-Living-Ecology Space has been analyzed. Taking Shangcang as an example, the elements and characteristics of its Production-Living-Ecology Space were dissected and summarized. The Characteristic Towns of Tianjin should make the best use of resources, improve rural infrastructure construction and public services, and promote the quality of ecological space.

Combined with the innovative teaching model of information technology and the concept of people-oriented as well as individualized instruction, this article discussed on the methods of inspiring and cultivating students’ interests and potential in learning and improving the self-regulated learning capability gradually, in order to realize the fundamental mission of education of cultivating virtues and morality and promoting students’ all-round development.

Study on the preparation of pullulan polysaccharide nanoparticles by dialysis

:The purpose of this experiment was to study the reaction of pullulan polysaccharide hydroxyl with phthalic anhydride, and to shrink the molecular group of pullulan polysaccharide by dialysis. The results showed that the configuration of the sugar group and the chemical position of the hydrogen could be determined by 1H-NMR; That is, the pullulan polysaccharide hydroxyl group had been combined with phthalic anhydride. According to the SEM, the particle shape was round. The mean particle sizes of 1∶3, 1∶6, 1∶9, 1∶12 were 119.2 nm, 146.5 nm, 176.4 nm and 114.3 nm, respectively.
