Soil Mechanics is a professional basic course. In the teaching process, taking the classroom as the carrier, teachers impart engineering scientific knowledge and carry out ideological and political education for students at the same time, which is a new subject of professional course teaching. Taking the course of Soil Mechanics as an example, sorting out the professional knowledge of the course, finding the meeting point between ideological and political education and professional education in combination with the development and integration of Ideological and political cases, designing the implementation scheme, exploring and practicing the construction of model courses, all help to realize the ideological and political education of the course, and achieve good results.

Introduction to Food Safety(Bilingual)is a specialized confined course for the major of food quality and safety. Under the background of national promotion of engineering education certification and improvement of engineering education quality, a new education and teaching system was explored based on “student-centered, results-oriented and continuous improvement” education concept via optimizing the teaching goals, teaching resources, teaching activities and evaluation system. It has achieved good teaching results and unanimous approval from teachers and students, providing reference for curriculum teaching reform to improve the quality of talent cultivation under the new situation.

Analysis of amino acid content in different cultivars of Vitis vinifera L. fruits

In order to study the differences of amino acid contentamong different cultivars of Vitis vinifera L. fruits ,six cultivars of Vitis vinifera L. in Changli County of Hebei Province were used as the main test materials .The amino acid contents in Vitis vinifera L. fruits were quantitatively analyzed by amino acid automatic analyzer. The results showed that the main amino acids in Vitis vinifera L. fruits were Glu, Arg, Pro and Asp. The total amount of amino acids in Vitis vinifera L. fruits was 368.33~197.17 mg/100g, and the content of essential amino acids was between 84.36~41.88 mg /100g, accounting for 17.88%~25.33% of the total amino acid content. Among the six cultivars, the difference of medicinal amino acid annonessenbial amino acid was significant. Glu was the most abundant amino acid among the 19 amino acids contained in the 6 varieties, among which the Glu content of Kyoho grape was 106.96 mg/100g, while the contents of Cys, Met and Tyr were few, which indicated that there were great differences in nutrition among different cultivars of Vitis vinifera L. fruits.

Compared with common Metal-Organic Framework materials(MOFs), UiO-66 has higher hydrothermal stability and chemical stability. UiO-66, UiO-66-NH 2 and UiO-66-(COOH) 2 samples were prepared by one-pot method, and then the carboxylated products were modified by PEI to prepare UiO-66-(COOH) 2-PEI samples. The UiO-66-NH 2 and UiO-66-(COOH) 2- PEI samples were subjected to fluorescence detection to determine the fluorescence intensity of the four metal salt solutions, such as aluminum chloride, zinc chloride, cobalt chloride, and lead chloride under the same concentration and different volumes of two samples. The samples and intermediate products were detected by infrared spectroscopy(IR). The results showed that the prepared UiO-66, UiO-66 intermediate and carboxylated UiO-66 product were successful. According to the results of fluorescence spectrophotometer, the above four metal salt solutions could enhance the fluorescence intensity of aminated UiO-66-(COOH) 2-PEI, and the volumes of metal salt solution were positively correlated with the enhancement effects. The above four metal salt solutions could quench the fluorescence intensity of UiO-66, and the volumes of metal salt solution were positively correlated with the quenching effects.

In view of the existing problems in the cultivation of professional core competence, according to the social demand for talent ability and starting from the concept of OBE, the “One body, two wings and three drives” type of food quality and safety professional core competence was constructed in this paper. “One body” referred to the main body of professional core competence, “whole-process control ability of food quality and safety”; “Two wings” referred to two important professional core competence “food quality control ability” and “food safety testing ability”; “Three drives” referred to the “food science” “professional quality” and “scientific” and “technological innovation” that is, the three aspects of knowledge, ability and quality as the driving force. From this, 2 first-level, 15 second-level and 15 third-level professional abilities were sorted out, professional core competence assessment system was developed and put into practice, and good results have been achieved.

Optimization of persimmon wine brewing process and analysis of aroma components

With persimmon as raw material, the effects of different fermentation temperature, initial acidity, initial sugar content and yeast inoculation amount on the quality of persimmon wine were studied through single factor and orthogonal experimental design. The brewing process of persimmon wine was optimized by measuring residual sugar, total acid, alcohol content and sensory score in persimmon wine. The results showed that the optimum fermentation conditions of persimmon wine were as follows: fermentation temperature was 20 ℃, initial acidity was 7.5 g/L, initial sugar was 200 g/L, and yeast inoculation amount was 0.2 g/L. Under this optimal combination, the sensory score of persimmon wine was 92 points. The aroma components of persimmon wine with the best technology were analyzed by GM-MS. The aroma components of persimmon wine included 68 volatile substances, including 15 alcohols, 27 lipids, 7 acids, 6 aldehyde and keone, 7 alkanes and 6 others. The main aroma ingredients of persimmon wine include: Isoamyl alcohol(apple brandy and spicy aroma), phenylpropanol(rose fragrance), isoamyl acetate(pleasant banana aroma), ethyl nonanoate(oil, fruit and brandy aroma), 2-methylbutyric acid(pleasant fruit aroma), and 3-hydroxy-2-butanone(milk aroma), etc.

In this paper, the discovery and classification, genome research, biological control and application of B. velezensis were reviewed in order to provide reference for further research and development.

Urea slow-release fertilizer is a new fertilizer to improve the utilization rate of nitrogen fertilizer, which is important for improving the utilization rate of nitrogen nutrients and crop yield. The application of sustained-release urea in the cultivation of grain crops such as wheat, corn, rice and economic crops such as cotton, soybean and so on in recent years is summarized in this paper. It is believed that the application of urea slow-release fertilizer in the process of crop planting has obvious effect of reducing nitrogen and increasing yield. At the same time, some problems still lacking research of urea sustained-release fertilizer in crop application are pointed out, such as the influence of slow-release urea fertilizer on the quality of crops, the influence of slow-release materials on environment and whether the slow-release curve matches the crop nutrient requirement curve. This study will provide some theoretical references for the future application research or widespread application of urea sustained-release fertilizer.

Since the 18th National Congress of the Communist Party of China, General Secretary Xi Jinping emphasized that moral education should run through the whole process of education, and that all kinds of courses should go together with ideological and political courses to form synergistic effect. Taking Hydraulics as an example, this paper made a preliminary exploration into the ideological and political teaching of specialized courses. The ideological and political elements contained in Hydraulics were deeply explored, and the ideological and political education was integrated into the classroom teaching of hydraulics from the aspects of China’s achievements in Hydraulics, scientists’ deeds, typical water conservancy projects, life practice, engineering ethics and classical culture. In a silent way, the students were taught the socialist core values, academic aspirations and professional ethics. The teaching mode and carrier approach of hydraulics course were innovated and improved the college student’s acquisition sense of ideological and political education.

In the process of ideological and political construction of Rural Sociology course, the author explored the path of adding ideological and political elements into the course construction: integrating ideological and political elements into the syllabus, teaching plan, teaching courseware and other teaching documents. In addition, according to the specific content of ideological and political elements in the classroom teaching, the author adopted different teaching methods, such as case method, comparative analysis method and heuristic teaching method. In the process of ideological and political construction of“rural sociology”course, the author also encountered some new problems to be solved in the subsequent teaching reform, including the systematic design of teaching and learning of the Rural Sociology course on the ideological and political education, the benign and effective interaction between teachers and students, and so on.

Phospholipase D (PLD) is a kind of enzyme which catalyzes the hydrolysis and base exchange of phosphodiesters. Functional phospholipids are prepared by transesterification of PLD. This paper introduced the molecular structure, catalytic mechanism and function of phospholipase. The production methods of phosphatidylserine (PS), phosphatidylglycerol (PG), phosphatidylethanolamine (PE) and phosphatidylinositol (PI) were mainly introduced, including physical extraction, chemical synthesis and enzymatic synthesis. Enzymatic synthesis is the most important method to prepare functional phospholipids. In addition, this paper prospected the future research on functional phospholipids. It is expected that the catalytic performance of PLD can be improved by improving the technical conditions and process system, which will provide theoretical and technical support for the industrial production of rare phospholipids.

Goldfish industry in Tianjin, which is one of the three goldfish headstreams in China, with more than three hundred years of history is an important part of Tianjin aquarium industry. Through an extensive investigation into practitioners of goldfish industry, the study collected the industry data and historical materials to describe the development history and development status of goldfish industry in Tianjin. The results indicated a fact of scale declining of goldfish keeping, the decline or even lose of traditionally dominant varieties and the aging of practitioner in goldfish industry of Tianjin. Finally, the method of PEST was used to analyse the factors that affected the development of goldfish industry in Tianjin to find corresponding strategies. The study aimed to provide ideas for the healthy development of the goldfish industry in Tianjin and references for the formulation of the industrial policies.

Ideological and political construction of college curriculum is a strategic measure to implement the fundamental task of moral education. Relying on the remote sensing elective course Looking at the Earth from Afar, Exploring the Changes of Universe and Earth, the ideological and political elements contained in “remote sensing” are fully explored and the abundant popular science forms were utilized to construct the teaching mode of “Ideological and Political Guidance & Science Popularization Drive” for remote sensing public elective courses. Taking advantage of the characteristics of wide range of students and flexible teaching methods in public elective courses, combined with the characteristics of unique remote sensing knowledge and strong application, this course adopted a scientific, novel, open and interactive mode to realize the imparting and cross-exploration of remote sensing technology knowledge, which provides a strong support for the inheritance of popular science. The combination of explicit knowledge education and implicit ideological and political education cultivated students' spirit of serving the majors and the grassroots, which was well received by the students. Students’ knowledge range and thinking of remote sensing have been significantly expanded, giving play to the educational advantages of public elective courses.

Optimization of quality improver and its effect on the storage characteristics of dried pork slice

Taking water content and water activity as evaluation indexes, the optimum proportion of sodium lactate, glycerin, sorbitol and trehalose in preserved pork was determined by single factor and L 9(3 4)orthogonal test. The dried pork slice was prepared according to the formula and then stored at 20 ℃ for 0, 15, 30, 45, 90, 120, 150 and 180 days. The water content, water activity, red value, total bacteria and mould number of the dried pork slice were determined, and the quality changes during storage were analyzed. The results showed that when 0.16% sodium lactate, 2% glycerol, 0.015% sorbitol and 1% trehalose were added in the processing of preserved pork, the moisture content of the product was 16.79%, and the water activity of the product was 0.677. During storage at 20 ℃, the number of moulds and total bacteria accorded with the national standard. The moisture content, water activity and red value gradually decreased. The shelf life of the product was 90 days, which was determined by sensory evaluation. At this time, the moisture content was 25.8%, the water activity was 0.778, and the red value was 19.8.

In order to study the effect of transport on dogs, Labrador retrievers were chosen and blood was collected before transport andafter 2-hour transport. Then serum cortisol, adrenocorticotropic hormone and biochemical indices were determined. The results showed: the concentration of cortisol and the activity of glutamic oxalacetic transaminase (AST) were significantly increased after transport ( P<0.05). The activity of Creatine kinase (CK) was significantly increased ( P<0.01). Adrenocorticotropic hormone (ACTH), alanine transaminase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), glucose (GLU) had no significant difference ( P>0.05). In conclusion, 2-hour transport stress can induce the change of serum cortisol, AST and CK of Labrador retriever.

In this study, minced beef bone(the mass ratio of bone to meat is 3∶7)was used as raw material. Traditional, spicy and fermented beef flavor were prepared by hot-pressing extraction and enzymatic hydrolysis followed by Maillard reaction with adding potassium sorbate in beef flavor as control group. The pH value, water activity and brightness of beef flavor were measured during storage at 4 ℃ and 20 ℃. The results showed that the pH values of BF with spices were different from those of other groups, and the changes of brightness and water activity were the same among the six groups. The shelf life of the beef flavor can reach 2.5 months and 40 d at 4 ℃ and 20 ℃, respectively. After modification, the product was stratified obviously.

In order to study the pathogenicity of early infection of Eimeria falciformis in mice, E. falciformis was invading by oral infection in the treatment group, while the control group was established with non-coccidia infection. Then the clinical symptoms, autopsy lesions, organ index, organ pathological histology and immunohistochemical characterization of cecum were observed for 36 hours to learn about the pathogenicity of early infection of E. falciformis in mice. The results showed that in the early stage of infection, E. falciformis maily damaged mucosal tissues of intestinal tract which caused degeneration, necrosis and shedding of epithelial and glandular epithelial cells, edema in strata submucosum, inflammatory cells infiltrating the mucosa but no visible pathological damage to other organs. According to immunohis chemical observation, the infections of mucosa and submucosa in the early stage were also observed. Hence, E. falciformis will be a candidate vaccine vector, which did not damage all the organs except cecum.

Profilin which was first discovered in 1976 was one of the earliest actin binding proteins, and has been widely studied because of its important biological role. Profilin1 is the most ubiquitous subtype because it can bind actin to regulate the polymerization of the cytoskeleton. Many recent studies have shown that Profilin1 also plays an important role in cancer and human diseases. This review mainly introduced the biological role of Profilin1, further explained its regulatory mechanism on the cytoskeleton, and discussed on the physiological and pathological role of Profilin1 in cancer and human diseases.

Physiological differences in saline-alkali tolerance of 8 North American begonia

In order to compare the physiological characteristics of different species of North American begonia on saline land in Tianjin, the soluble protein, soluble sugar, free proline, malondialdehyde content, membrane permeability, SOD, POD enzyme activity and other physiological and biochemical indexes in leaves were determined, with eight species of North American begonia as test material in Jinghai. The results showed that the highest soluble protein content was in Kells(42.235 mg/g), the lowest was in Donghong(14.763 mg/g); the highest soluble sugar content was in Kells(0.163%)and Julie(0.151%), the lowest was in Wumei(0.103%); the highest free proline content was in Kells(7.285 μg/g)and Donghong(6.211 μg/g), the lowest was in Royal(2.747 μg/g). The highest malondialdehyde content was in Kells(0.133umol/g), the lowest was in Adam(0.079 μmol/g)and Julie(0.075 μmol/g); the highest membrane permeability(34.96%)was in Donghong, the lowest was in royal(8.22%); the highest POD enzyme activities was in Xiyangyang(50.771 U/g·min)and Wumei(54.254 U/g·min), the lowest was in Donghong(12.208 U/g·min); and there was no difference in SOD activity among the eight species of North American Begonia, and the enzyme activity was between 0.512 U/g and 0.613 U/g. According to the comprehensive analysis of membership functions, their order of saline tolerance were Kells>Donghong>Ruby>Julie>Adam>Royal>Xiyangyang>Wumei.

To affirm the optimized chemical constitution of submerged fermentation culture medium for Monascus exopolysaccharide(EPS), the experiment was carried out with rice powder as the main substrate medium and by single factor, orthogonal and other experimental methods. The result shows that the best supplementary nitrogen source is soybean powder added to 10 mg/mL. The yield of EPS was inhibited by MnSO 4, CaCl 2, CuSO 4, FeCl 3, FeCl 2, ZnSO 4, and activated by KCl, KH 2PO 4, NaCl. In addition, all of these inorganic salts’ amount added are 0.15 mg/mL. Consequently, the fermentation mediums were optimized as follows: the powder of rice and the powder of soybean each account for 40 mg/mL and 10 mg/mL of the total amount. The dosages of KCl, KH 2PO 4 and NaCl were all 0.15 mg/mL. Under this condition, the yield of EPS reached 112.3 mg/mL on average, after culture age of 120 h. It is about twice the yield of liquid fermented polysaccharides based on rice flour alone.

Bladder calculi and pyometra are common clinical cases of pets with high morbidity. Dog calculi is a stone in the urinary system due to various reasons. Dog stones include kidney, ureter, bladder and urethral stones, among which bladder and urethral calculi are the most common especially. A stone can cause a pet to urinate badly, no urine, abdominal pain, and severe urine leakage in the abdominal cavity, causing peritonitis. Pyometra is due to endometrial hyperplasia, the existence of a purulent material in the uterus accumulation of fluid. It is easy to increase the incidence rate and mortality rate of pyometra after uterus infection. However, there is only few cases when both bladder calculi and pyometra occur in one dog. This report followed up cases of 8-year-old pekingese and 7-year-old poodles suffering both from bladder calculi and pyometra. After clinical diagnosis, such as blood routine examination, biochemical examination, X-ray examination and b-mode ultrasound examination, they were finally diagnosed as the complication of bladder calculi and pyometra. Two cases had bladder calculi removed and uterus removed by surgery. Through drug treatment and postoperative care, spirit of the wound recovered with normal eating and micturition recovery after a week. The stitches were removed 10 days later and the wound healed well. After the clinical symptoms of the disease were analyzed, the systematic research report on the professional clinical examination, detailed treatment and nursing can provide clinical reference for the treatment of bladder calculi and uterus suppurative disease.

Mesotrione is an important herbicide in maize field, and one of its main degradation pathways is microbial degradation. HF/6-31G(d)was used to perform the structural optimization and frequency analysis of mesotrione. The possible oxidation and reduction reactions in micro-organisms of the compound were studied. The entropy and Gibbs free energy of different redox processes were compared. The calculation results showed that mesotrione will give priority to produce pro6 with the catalytic effect of enzymes in microorganisms, which helps us to further understand the degradation behavior of mesotrione by microorganisms in environment.

Nicotinamide is a kind of pyridine derivative, which has various pharmacological effects such as inhibition of melanic transmission, anti-free radical, anti-aging, anti-inflammatory and enhancement of skin barrier after transdermal administration. However, due to its structure of amide, nicotinamide could be oxidized to nicotinic acid, which could cause dermal irritation and side effects. Thus, water-in-oil(W/O)ointment was prepared to enhance stability and compliance of nicotinamide. In this investigation, comprehensive scoring of homogeneity, extensibility, centrifugal stability and cold/heat resistance was set as the evaluation indicator to obtain the optimal proportion of oil phase, emulsifier and coemulsifier through single-factor experiment and orthogonal screening. under the optimal formulation. Span-60 and Tween-80 were used as emulsifier and coemulsifier respectively with the mass percentage of 1%, and the weight ratio of oil to water was 6∶4. Under the selected formulation, storage stability of ointment remained in 3 months under 25 ℃, which provides the basis for practical application.

Effects of pH on chlorophyll fluorescence parameters and growth of Chlorella vulgaris

To determine the optimum pH conditions and provide basic information for intensive production of Chlorella vulgaris, the effects of different pH values on chlorophyll fluorescence, chlorophyll content and cell density during its culture process have been studied. The results show that when pH is 9 the maximum energy conversion rate( Fv/ Fm), the actual photochemical efficiency( ΦPSⅡ)and the quantum efficiency( Yield)decreased; meanwhile, the potential energy( Fv/ F0), relative electronic conversion rate( ETR), chlorophyll content( CHL)and the density of Chlorella increasing trend was minimum. When pH is 7, chlorophyll fluorescence are higher than the other groups with the values 1 613.05 µg/L, 1.13×10 7 cells/mL respectively. The chlorophyll content and cell density of pH 9 treatment group were 883.82 µg/L and 6.77×10 6cells/mL. The optimum growth pH of Chlorella is 7, and pH 9 can obviously inhibit the growth of Chlorella.

:To study the molecular epidemiology of Cyniclomyces guttulatus and its pathogenicity to dog intestine, 222 fecal samples were collected from three large police dogs bases in Tianjin area. Microscope examination, separated culture, dyeing identification, yeast genome extraction and 18S rRNA gene identification of C.guttulatus in the fecal samples were carried out, and the isolated strains were studied for the pathogenicity of the dogs. Results showed that there were 35 C.guttulatus positive samples from the police dog base in Xiqing, and the positive rate was about 15.77%. The proliferation mode of the yeast was identified as germ producing yeast by dyeing. The target band was amplified by 18S rRNA gene to get about 400 bp, and 6 experimental dogs were studied for the pathogenicity. The symptoms of diarrhea and stool were found in two dogs. By fecal microscopic examination droppings of yeast or yeast spore were found. Therefore, the C.guttulatus may be as an opportunistic pathogenic bacteria, which can cause intestinal infection under certain conditions in dogs.

In order to clarify the associated pathogens of companion animal suppurative diseases, uterine secretions from a patient pyometra dog at animal hospital affiliated to the college were collected sterilely for pathogen isolation and identification. The isolate was identified as Citrobacter freundii by bacterial isolation and culture, colony morphology observation and Gram’s staining observation, biochemical test and 16S rRNA gene analysis. Antibiotic susceptibility results showed that the isolated bacterium is resistant to Maddie and ampicillin, moderately sensitive to aztreonam, cefazolin, and cefoxitin, highly sensitive to cefoperazone, ceftriaxone, cefepime and other antibiotics, providing reference for prevention and related diseases control of companion animals.

It is assumed that the infiltration process is divided into two stages of pre-infiltration and post-infiltration. The ratio of soil saturated zone to unsaturated zone is a constant 2 β in the pre-infiltration stage, and the thickness of unsaturated zone increases with infiltration. The thickness of unsaturated zone in the post-infiltration stage no longer changes with time, which is a fixed value,and only the thickness of saturated zone changes with time. Based on this, Green-Ampt model is improved. The traditional Green-Ampt model and improved model were used to fit six groups of infiltration test data. The correlation coefficient of traditional model fitting was 0.982(the average of six groups of data). Compared with the traditional model, the fitting accuracy of the improved model was obvious. The correlation coefficient was increased by 0.016, reaching 0.998 (corresponding to β=15). The data of two groups were especially effective, which were increased by 0.07 and 0.04 respectively, both reaching over 0.99. The improved model has the same strong physical meaning as the traditional model. However, compared with the traditional model, the stable infiltration rate obtained by the improved model is closer to the test value and the improved model has wider applicability.

In order to clarify the causes of the fester disease in companion animals, the study involved isolation, identification and drug susceptibility analysis of pathogenic bacteria from an 11-year-old female dog’s uterus who were admitted to the veterinary hospital of Tianjin Agricultural University. According to the results of colony morphology, smear staining microscopy, physiological and biochemical reaction and 16S rRNA gene analysis, the bacteria was identified as a Bacillus pyocyaners. Susceptibility analysis showed that the isolate is highly sensitive to aztreonam, cefdinir, cefotaxime, cefepime, ceftazidime, ofloxacin, streptomycin, kanamycin, piperacillin and tobramycin. Moreover, this isolate is moderately sensitive to ceftriaxone, cefuroxime and cefoxitin, but resistant to ampicillin, cefazolin, cefazine and medemycin. The research results provided references for controlling infection caused by Bacillus pyocyaners.
