Induction of polyploid Robinia pseudoacacia L. and analysis of anatomical structure of leaves

Abstract: Tetraploid Robinia pseudoacacia L. has been widely planted because of its strong adaptability and large biomass. In this study, the seedlings of R. pseudoacacia were treated with 5 different concentrations colchicine to induce tetraploid, and the changes of the anatomical structure of the tetraploid leaves after induction were studied. It was found that 0.05% colchicine could achieve better induction effect in one day. The thickness of the upper epidermis, the palisade tissue, sponge tissue and leaf thickness of the tetraploid R. pseudoacacia leaves were significantly higher than those of diploid, and the cell tense ratio of tetraploid ones were significantly improved, which indicated that R. pseudoacacia had strong resistance to stress after tetraploidization. The results can provide reliable technical support for the polyploid breeding of R. pseudoacacia, provide reference and theoretical support for the ploidy breeding of R. pseudoacacia, and provide morphological and anatomical basis for explaining the formation principle of the resistance of tetraploid R. pseudoacacia.
