With the development of nano-medicine, a novel kind of self-assembled nanoparticle was fabricated through electrostatic crosslink method using negative-charged lecithin and positive-charged chitosan. This kind of self-assembled nano-carrier could be used to improve oral bioavailability of drugs with poor solubility. In this study, we utilized this nano-carrier to facilitate the drug delivery of berberine hydrochloride, whose water solubility is low to 1:200(g:mL). At the same time, the method for entrapment efficiency( EE)measurement was established. Afterwards, orthogonal screening was used to determine the optimal formulation of drug-loaded self-assembled nanoparticle, in which entrapment efficiency( EE)was set as evaluation indicator. In the selected formulation, pH value of water phase was 8.0, volume ratio of ethanol phase/water phase was 8:92, and mass ratio of lecithin/chitosan was 20:1. Three batches self-assembled nanoparticle were prepared under the selected formulation, and evaluation results showed that the average particle size, Zeta potential and entrapment efficiency was(214.8±9.3)nm,(24.3±3.1)mV and(82.4±5.2)%, separately. Berberine hydrochloride loaded self-assembled nanoparticle remains high stability under 4 ℃ storage, which provided the basis for practical application.

Bladder calculi and pyometra are common clinical cases of pets with high morbidity. Dog calculi is a stone in the urinary system due to various reasons. Dog stones include kidney, ureter, bladder and urethral stones, among which bladder and urethral calculi are the most common especially. A stone can cause a pet to urinate badly, no urine, abdominal pain, and severe urine leakage in the abdominal cavity, causing peritonitis. Pyometra is due to endometrial hyperplasia, the existence of a purulent material in the uterus accumulation of fluid. It is easy to increase the incidence rate and mortality rate of pyometra after uterus infection. However, there is only few cases when both bladder calculi and pyometra occur in one dog. This report followed up cases of 8-year-old pekingese and 7-year-old poodles suffering both from bladder calculi and pyometra. After clinical diagnosis, such as blood routine examination, biochemical examination, X-ray examination and b-mode ultrasound examination, they were finally diagnosed as the complication of bladder calculi and pyometra. Two cases had bladder calculi removed and uterus removed by surgery. Through drug treatment and postoperative care, spirit of the wound recovered with normal eating and micturition recovery after a week. The stitches were removed 10 days later and the wound healed well. After the clinical symptoms of the disease were analyzed, the systematic research report on the professional clinical examination, detailed treatment and nursing can provide clinical reference for the treatment of bladder calculi and uterus suppurative disease.

At the same time, orthogonal screening was used in formulation selection in order to obtain stable ointment formulation with preferred skin-feeling. In this investigation, flavonoid ingredients were acquired through ultrasonic extraction firstly. Afterwards, comprehensive scoring of homogeneity, extensibility, centrifugal stability and cold/heat resistance was set as the evaluation indicator to obtain the optimal proportion of oil phase, emulsifier and coemulsifier. In this ointment formulation, Span-60 and Tween-80 were used as emulsifier and coemulsifier respectively, and lanolin/vaselin/stearic were used as muti-compositional oil phase with quality ratio of 1∶3∶4. After prepared under the optimal formulation, in which the mass percentage of oil phase, Span-60 and Tween-80 were 35.0%, 4.0% and 4.5% respectively, stable ointment was acquired in oil-in-water type. This study provides the basis for further application of Radix angelicae sinensis extracts ointment.

The method for rapid detection of oxytetracycline residues in milk was established by combining mid-infrared (MIR) spectroscopy with partial least squares discriminant analysis(PLS-DA). Firstly, 62 pure milk samples and 62 milk samples adulterated with different concentrations of oxytetracycline(0.02-30.00 mg/L)were prepared respectively. The MIR spectra of all samples were collected in the range of 400-4 000 cm -1. After the pretreatment of original spectral data, the PLS-DA models were established in the full band and sub band ranges, respectively. The results show that different wave-number ranges had a great influence on the prediction model of milk adulterated with oxytetracycline. The classification accuracies of the constructed PLS-DA models were 100% for calibration set in the full band and sub band ranges, and the correlation coefficients of models were between 0.995 and 0.997 for prediction set, which shows that the models had a good fitting effect. The classification accuracies of constructed models in the full band and in the range of 2 000-4 000 cm -1 were 90.0% for prediction set.
