Amination modification and characterization of UiO66

Compared with common Metal-Organic Framework materials(MOFs), UiO-66 has higher hydrothermal stability and chemical stability. UiO-66, UiO-66-NH 2 and UiO-66-(COOH) 2 samples were prepared by one-pot method, and then the carboxylated products were modified by PEI to prepare UiO-66-(COOH) 2-PEI samples. The UiO-66-NH 2 and UiO-66-(COOH) 2- PEI samples were subjected to fluorescence detection to determine the fluorescence intensity of the four metal salt solutions, such as aluminum chloride, zinc chloride, cobalt chloride, and lead chloride under the same concentration and different volumes of two samples. The samples and intermediate products were detected by infrared spectroscopy(IR). The results showed that the prepared UiO-66, UiO-66 intermediate and carboxylated UiO-66 product were successful. According to the results of fluorescence spectrophotometer, the above four metal salt solutions could enhance the fluorescence intensity of aminated UiO-66-(COOH) 2-PEI, and the volumes of metal salt solution were positively correlated with the enhancement effects. The above four metal salt solutions could quench the fluorescence intensity of UiO-66, and the volumes of metal salt solution were positively correlated with the quenching effects.

In view of the existing problems in the cultivation of professional core competence, according to the social demand for talent ability and starting from the concept of OBE, the “One body, two wings and three drives” type of food quality and safety professional core competence was constructed in this paper. “One body” referred to the main body of professional core competence, “whole-process control ability of food quality and safety”; “Two wings” referred to two important professional core competence “food quality control ability” and “food safety testing ability”; “Three drives” referred to the “food science” “professional quality” and “scientific” and “technological innovation” that is, the three aspects of knowledge, ability and quality as the driving force. From this, 2 first-level, 15 second-level and 15 third-level professional abilities were sorted out, professional core competence assessment system was developed and put into practice, and good results have been achieved.

Bacillus velezensis, which is a new species of Bacillus, can promote plant growth and inhibit plant pathogens in agriculture and has great potential to be developed as a biocontrol agent. It can also be used in industry to degrade industrial harmful by-products and be widely used in biomedicine.Therefore, it has attracted much attention in recent years. In this paper, the discovery and classification, genome research, biological control and application of B. velezensis were reviewed in order to provide reference for further research and development.

Optimization of persimmon wine brewing process and analysis of aroma components

With persimmon as raw material, the effects of different fermentation temperature, initial acidity, initial sugar content and yeast inoculation amount on the quality of persimmon wine were studied through single factor and orthogonal experimental design. The brewing process of persimmon wine was optimized by measuring residual sugar, total acid, alcohol content and sensory score in persimmon wine. The results showed that the optimum fermentation conditions of persimmon wine were as follows: fermentation temperature was 20 ℃, initial acidity was 7.5 g/L, initial sugar was 200 g/L, and yeast inoculation amount was 0.2 g/L. Under this optimal combination, the sensory score of persimmon wine was 92 points. The aroma components of persimmon wine with the best technology were analyzed by GM-MS. The aroma components of persimmon wine included 68 volatile substances, including 15 alcohols, 27 lipids, 7 acids, 6 aldehyde and keone, 7 alkanes and 6 others. The main aroma ingredients of persimmon wine include: Isoamyl alcohol(apple brandy and spicy aroma), phenylpropanol(rose fragrance), isoamyl acetate(pleasant banana aroma), ethyl nonanoate(oil, fruit and brandy aroma), 2-methylbutyric acid(pleasant fruit aroma), and 3-hydroxy-2-butanone(milk aroma), etc.

Ideological and political construction of college curriculum is a strategic measure to implement the fundamental task of moral education. Relying on the remote sensing elective course Looking at the Earth from Afar, Exploring the Changes of Universe and Earth, the ideological and political elements contained in “remote sensing” are fully explored and the abundant popular science forms were utilized to construct the teaching mode of “Ideological and Political Guidance & Science Popularization Drive” for remote sensing public elective courses. Taking advantage of the characteristics of wide range of students and flexible teaching methods in public elective courses, combined with the characteristics of unique remote sensing knowledge and strong application, this course adopted a scientific, novel, open and interactive mode to realize the imparting and cross-exploration of remote sensing technology knowledge, which provides a strong support for the inheritance of popular science. The combination of explicit knowledge education and implicit ideological and political education cultivated students' spirit of serving the majors and the grassroots, which was well received by the students. Students’ knowledge range and thinking of remote sensing have been significantly expanded, giving play to the educational advantages of public elective courses.

Profilin which was first discovered in 1976 was one of the earliest actin binding proteins, and has been widely studied because of its important biological role. Profilin1 is the most ubiquitous subtype because it can bind actin to regulate the polymerization of the cytoskeleton. Many recent studies have shown that Profilin1 also plays an important role in cancer and human diseases. This review mainly introduced the biological role of Profilin1, further explained its regulatory mechanism on the cytoskeleton, and discussed on the physiological and pathological role of Profilin1 in cancer and human diseases.

Progress in application of fluorescence spectroscopy for detection of PAHs in soil

Polycyclic aromatic hydrocarbons(PAHs)are a kind of toxic, carcinogenic, hard-to-degrade persistent organic pollutants, 90% of PAHs in the environment are finally enriched in the soil, and soil PAHs pollution is particularly prominent. Therefore, the realization of rapid and on-site detection of PAHs in soil is practically significant for soil pollution investigation, environmental management, production and life. Compared with traditional detection methods, fluorescence spectrometry has many advantages such as good selectivity, high sensitivity and low detection limit, and has been widely used for the detection of PAHs in soil. The article reviewd the application and research progress of conventional fluorescence spectroscopy, three- dimensional fluorescence spectroscopy, two-dimensional fluorescence correlation spectroscopy and laser-induced fluorescence spectroscopy for the direct detection of PAHs in soil in recent years. The application of fluorescence spectroscopy is discussed in the extraction of PAHs characteristic information, quantitative analysis, the influence of soil physical and chemical properties on PAHs fluorescence intensity and correction, etc. and the prospect were discussed to provide reference for further realization of fluorescence spectroscopy technique for direct deaction of PAHs in soil.

Study on the estimation model of summer maize leaf SPAD value based on hyperspectrum

Quick and accurate estimation of SPAD values of summer maize leaves can provide data support for monitoring the growth of summer maize in a large area. The spectral reflectance of summer maize leaves at different growth stages, such as jointing stage, tasseling stage, filling stage and milky stage, was simulated by PROSPECT model, the modeling method of estimating SPAD value based on hyperspectral technique. The results showed that:(1)the optimal hyperspectral index estimation model for SPAD value was proposed for each growth period, and the corresponding optimal hyperspectral indexes were REIP, mND705, mND705, and G-M. The decision of the model coefficients( R 2)are: 0.625 9, 0.691 2, 0.591 9 and 0.579 9, the root mean square error( RMSE)are: 14.4, 12.3, 6.3 and 9.4, and the relative error( RE)are: 29.1%, 22.1%, 9.3% and 16.9%.(2)A general hyperspectral index mND705 model was proposed to estimate SPAD value for the whole growth period. The R 2 of the model were 0.597 3, 0.691 2, 0.591 9 and 0.402 4, the RMSE were 12.5, 12.0, 6.4 and 7.3, and the RE were 24.2%, 21.5%, 10.4% and 11.8%, respectively. Through comparative analysis, it is concluded that the general model R 2 is high and RE is low. Therefore, the general model proposed in this paper has better estimation ability and high precision for the inversion of SPAD value of summer maize leaves.

Urea slow-release fertilizer is a new fertilizer to improve the utilization rate of nitrogen fertilizer, which is important for improving the utilization rate of nitrogen nutrients and crop yield. The application of sustained-release urea in the cultivation of grain crops such as wheat, corn, rice and economic crops such as cotton, soybean and so on in recent years is summarized in this paper. It is believed that the application of urea slow-release fertilizer in the process of crop planting has obvious effect of reducing nitrogen and increasing yield. At the same time, some problems still lacking research of urea sustained-release fertilizer in crop application are pointed out, such as the influence of slow-release urea fertilizer on the quality of crops, the influence of slow-release materials on environment and whether the slow-release curve matches the crop nutrient requirement curve. This study will provide some theoretical references for the future application research or widespread application of urea sustained-release fertilizer.

Flipped classroom is a new teaching concept and method, emphasizing “student-centered”, flipping the traditional teaching process, putting students’ learning process of new knowledge before class, and using class time for discussion, sharing and questioning. The implementation of flipped classroom is conducive to enhancing students’ participation in learning and enhancing their autonomous learning ability. Flipped classroom changes the relationship between teachers and students, and students become the leading role and creator of knowledge. Based on the explanation for the theory of flipped classroom, the paper analyzed the application of flipped classroom in the teaching of logistics management. The process of teaching was designed into four stages including the design of teaching objectives and contents, self-learning before class, in class teaching, consolidation and improvement after class; and countermeasures to improve the teaching effect of flipped classroom were put forward, including considering the differences of students, changing the teaching philosophy of teachers, and improving the information service system of education.

To understand the current situation and development trend of black soldier fly research, the literature and patent achievements from 1990 to 2020 were searched and analyzed based on CNKI and Baiteng database. The results showed that there was a trend of explosive growth in the number of related literature about black soldier fly since 2015, and that will continue to increase in the future. In the existing literature, the research work mainly focused on the growth and development, waste disposal, insect protein, medicine research and development of black soldier fly. The research covered a wide range of subjects, focusing on the environment and agricultural biology. The number of utility model patents published in China was much higher than those of invention authorization and appearance design, and the number of utility model patents reached the peak in 2019. Research funding in this field needs to be further strengthened.

Study on the effect of different fermentation methods on volatile aroma components in persimmon wine

The yield of persimmons in Tianjin is relatively large, and a lot of persimmons are wasted every year. The development of persimmon wine can effectively avoid the waste of persimmons. At present, persimmon wine is generally directly fermented with fruit pulp, but different processes of fermentation have a great influence on the formation of aroma components of persimmon wine. Therefore, this paper uses electronic nose, gas-mass spectrometry and sensory evaluation to compare and analyze the aroma components of persimmon wine with different fermentation methods from different angles. The test results showed that the number of lipids in the volatile aroma components of persimmon wine fermented with clear juice was 27, followed by pulp fermentation; the comprehensive sensory score of clear juice fermentation was higher than that of pulp fermentation and peel fermentation, which was detected by gas-mass spectrometry. Persimmons fermented with clear juice have the same result with more aroma components. Through the principal component analysis of the gas-mass spectrometry detection results, the main aroma substances and contents of the three persimmon wines were obtained. The persimmon wine fermented with clear juice is more aromatic, making the persimmon wine more typical.

To achieve an effective connection between the two, there are still difficult problems such as difficulty in following up and changing ideas, difficulty in improving and upgrading the system and mechanism, and difficulty in fully stimulating the endogenous power. Screening of tomato callus and acquisition of stress resistant mutants

Tomato petioles were used as explants to establish tomato callus regeneration system, then the mutant cells of tomato callus were screened by PEG-8000, and the regeneration of callus cells was induced at the same time, so as to screen regenerated plants with strong growth potential and stress resistance. The results showed that the callus with hard texture and loose structure could be induced on the medium of MS + 1.0 mg/L BA + 2.0 mg/L 2,4-D ; and the highest regeneration rate was 100% when the callus was transferred to the medium of MS + 1.0 mg/L BA; the regenerated plantlets with fast growth, robust and stress-resistant characteristics could be obtained by inoculating the callus on the regeneration medium containing PEG-8000, and the ideal screening concentration of PEG-8000 was 5%. In this study, stress-resistant regenerated plants were obtained by directional screening of callus mutant cells, which could provide reference for plant stress-resistant breeding.

Cloning and structure prediction of cell division cyclin 42 gene from Cynoglossus semilaevis

Cdc42 is a subclass of Rho superfamily enzymes that regulates cytoskeleton and actin activity, and participates in immune and inflammatory responses. In this study, the specific primers were designed for Cdc42 gene( CsCdc42)of Cynogl ossus semilaevis, and its cloning and structure prediction were performed by RT-PCR and bioinformatics methods. The results showed that the fragment of CsCdc42 gene was 847 bp in length, encoding 191 amino acids(aa), and contained 7 epitopes (27-28, 61-63, 90-92, 120-122, 131-133, 164-166 and 181-183 aa), one RHO domain(6-179 aa), ATP/GTP binding site and acylation site, respectively. The protein structure was predicted by helix(30.9%), β-fold(24.6%)and irregular coil(44.5%). The results of sequence alignment showed that the homology of amino acid sequences of CsCdc42 was 99.43%, 97.91%, 95.81%, 94.76% and 91.10%, respectively, for zebrafish( Danio rerio), medaka( Oryzias latipes), rainbow trout( Oncorhynchus mykiss), turbot ( Scophthalmus maximus)and shrimp( Penaeus vannamei). Phylogenetic tree analysis showed that Cdc42 was closely related to zebrafish( Danio rerio), human( Homo sapiens)and several marine fishes, and naturally grouped into one cluster. These results provide a scientific reference for further study of the function of cyclin-related genes in Cynoglossus semilaevis.

Study on extraction optimization of polyphenols from Mesembryanthemum crystallinum and its antioxidant and bacteriostatic activities

In this study, the extraction method of polyphenols from Mesembryanthemum crystallinum was optimized, and the antioxidant capacity and antibacterial activity of polyphenols from M. crystallinum were determined. The results showed that the optimal extraction conditions were as follows: ethanol concentration was 60%, solid-liquid ratio was 1∶10(g: mL), extraction temperature was 80 ℃, extraction time was 120 min, under which the extraction rate of polyphenols was 1.42%. The scavenging rates of polyphenols from M. crystallinum to 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)and hydroxyl radicals increased with the increase of polyphenols concentration. When the concentration of polyphenol extract was 0.10 mg/mL, the scavenging rates of DPPH and hydroxyl radicals were 64.19% and 88.10%, respectively. The inhibitory effect of polyphenol extract on Escherichia coli was obvious. When the extract concentration was 3 mg/mL, the diameter of inhibition zone was 12 mm.

Effects of different DNA extraction methods on structural analysis of intestinal microbiota from Cynoglossus semilaevis based on high-throughput sequencing

The purpose of this study was to analyze the effects of different DNA extraction methods on structural analysis of intestinal microbiota from C. semilaevis based on high throughput sequencing and comprehensively learn about the structural characteristics of its intestinal flora. In this study, three DNA extraction methods including combination of lysozyme and ultrasonic lysis(CLU), Zirmil-beating cell disruption(ZBC)and QIA commercial kits were used to extract the genomic DNA of intestinal microflora from healthy C. semilaevis. The DNA samples were sequenced on an Illumina MiSeq high-throughput sequencing platform, and the structural characteristics and diversities of the intestinal microflora were analyzed. The results showed that a total of 7 516 OTUs were obtained across all samples extracted by the three DNA extraction methods. The identified dominant bacterial taxa were similar among these samples. At Phylum level, the dominant bacterial taxa included Proteobacteria, Firmicutes, Spirochaetae and Bacteroides. Through Alpha diversity index analysis, the corresponding Chao1 indexes of CLU, ZBC and QIA methods were 5 668, 5 868 and 4 090 respectively, and the Simpson index were 0.021, 0.022 and 0.018 respectively. The Chao1 and Simpson indexes of QIA methods were all lower than those of other methods. Beta diversity analysis based on the UniFrac distance matrix showed that the microbial community structure and species richness were more similar between the CLU and QIA samples. These results indicated that these three different DNA extraction methods reached the similar results in the major dominant intestinal microflora, based on the investigation using high-throughput sequencing analysis. However, biases occurred in the identification results of some less abundant intestinal microflora. The marine shellfish aquaculture industry has the highest yield in the marine aquaculture industry of circum-Yellow- Sea. However, the huge economic incentives in recent years have led to the high density of marine shellfish culture in the coastal waters, and the recent large-scale construction of marine ranches in the coastal waters of circum-Yellow-Sea has resulted in serious shortage of microalgae for the survival and growth of marine shellfish. Scarcity and high-intensity grazing pressure induced the occurrence of brown tide disasters caused by algae species, and the irregular outbreaks of carapace, ascidians, echinoderms and other marine shellfish hostile organisms lead to the reduction of shellfish in coastal waters. Incidents such as reduced production and no harvest have occurred from time to time, seriously jeopardizing the sustainable and healthy development of the marine shellfish aquaculture industry of circum-Yellow-Sea. The author has initially established targeted emergency response measures for ecological disasters in marine shellfish aquaculture, and scientifically put forward countermeasures and suggestions to ensure the healthy development of marine shellfish aquaculture in circum-Yellow-Sea.

Effects of salicylic acid on physiological characteristics of alfalfa seedlings under salt stress

In order to understand the effects of exogenous salicylic acid on physiological characteristics of alfalfa seedlings under salt stress, the contents of soluble protein, soluble sugar, proline and malondialdehyde, as well as the activities of peroxidase and superoxide dismutase were analyzed by soaking the alfalfa variety ‘Zhongmu No. 3’ in 1 mmol/L salicylic acid solution for 12 h and 5 kinds of salt contents. The results showed that the soluble protein content, soluble sugar content, superoxide dismutase activity and peroxidase activity of alfalfa seedlings were significantly increased compared with the control group by salicylic acid in the range of soil salt content from 0.20% to 0.52%. The proline content of alfalfa seedlings was significantly increased with salicylic acid at soil salt content from 0.35% to 0.52%. The content of malondialdehyde in alfalfa seedlings was significantly reduced by salicylic acid when the soil salt content was 0.20%~0.52%. The salt resistance of alfalfa seedlings could be enhanced with exogenous salicylic acid by enhancing the activity of antioxidant enzymes, increasing the content of osmotic substances and decreasing the content of malondialdehyde.

Effects of potassium fertilization on the quality of ‘Muscat Hamburg’ grapes

In order to explore the effects of potassium fertilizer on the quality of ‘Muscat Hamburg’ grapes, the effects of three potassium fertilization(30, 40 and 50 kg/667m 2)on fruit quality(hardness, fruit shape index, water content, soluble solids, soluble sugar, titratable acid, soluble protein, and VC content)were studied using 6-year-old ‘Muscat Hamburg’ grapes as the test material. The results showed that when the potassium fertilization was 30 kg/667m 2, the hardness(1.03 kg/cm 2), soluble sugar content(5.37 mg/g)and fruit shape index(1.2)of the fruit was the highest, which was significantly higher than other treatments and its titratable acid content(0.22%)and the water content(74.35%)were significantly lower than other treatments. The comprehensive analysis of the membership function showed that the potassium fertilization treatment 30 kg/667m 2 had the best comprehensive quality of the fruit, with an average membership function value of 1.16, followed by the treatment of 40 kg/667m 2, with an average membership function value of 0.80. Therefore, supplementary application of 30 kg/667m 2 potassium fertilization can significantly improve the quality of ‘Muscat Hamburg’ grapes.

To investigate the effect of myostatin( MSTN)on skeletal muscle fat metabolism, MSTN -/- Mongolian cattle (MG.ZJY)and wild Mongolian cattle(MG.DZ), MSTN -/- Luxi cattle(LX.ZJY)and wild Luxi cattle(LX.DZ)and MSTN -/- Mongolian Black Cattle(HN.ZJY)and wild Mongolian black cattle(HN.DZ)leg gluteal muscle tissue as a sample, quantitative proteomics and phosphorus Acidified proteomics technology were used to screen differential proteins and differentially phosphorylated proteins. These differential proteins and phosphorylated proteins were statistically analyzed by online bioinformatics analysis software and website, and triglyceride content in various tissues were detected. The results showed that the identified multiple differential proteins were all related to fat metabolism, and these differential proteins were mainly enriched in fatty acid degradation, fatty acid metabolism and PPAR signaling pathways. Tissue triglyceride assay showed that the triglyceride content in muscle tissue of MSTN -/- cattle was significantly decreased compared with that of wild cattle. The results of this study showed that knockout of MSTN gene could promote fatty acid metabolism in bovine skeletal muscle. The results lay a foundation for further research on the effect of MSTN gene on skeletal muscle metabolic mechanism.

The 48-h LC 50 value of Galla Chinansis to Moina macrocopa was studied by acute toxicity test. The influence of Galla chinensis(1.25, 2.50, 5.00, 10.00, 20.00, 40.00 mg/L)on the statistical parameters of life table of Moina macrocopa was studied by means of life table test. The results showed that Chinese herbal medicine Galla Chinansis had significant effect on survival rate and reproduction rate of Moina macrocopaP<0.05), and the 48-h LC 50 of Galla Chinensis on Moina macrocopa was 79.57 mg/L. The life table parameters showed that the population intrinsic growth rate and net reproductive rate were significantly decreased( P<0.05)at high concentration(40 mg/L), and the values were [(0.015 3±0.007 6)h -1] and [(1±0.67)ind.] respectively.

To investigate the clinical treatment and prognosis of bladder calculi complicated with cystomyoma in dogs,a case of senile spotted dog bladder calculi was treated in a pet hospital. In the course of surgical treatment of bladder calculi in an old dalmatian dog, there was an approximately 20 cm×15 cm×15 cm uneven surface neoplasm attached to the dorsal side of the bladder body. The bladder mucosa at the junction with the neoplasm was found to be intact, the bladder serous membrane surface was closely connected with the neoplasm, and the surface of the neoplasm had a complete membrane structure. Histopathological diagnosis showed that the tumor mass was dominated by proliferating spindle cells and was a benign Leiomyoma of the bladder. After cystolithotomy and surgical removal of the tumor, the dog recovered well.

Behavioral ecology is a discipline that studies the evolutionary basis of animal behavior due to ecological pressure. After studying the four problems that need to be solved and the direct causes, ontogeny, survival value and systematics of animal behavior, behavioral ecology emerges from ethology. Studying the causes and laws of these behaviors can help us better understand self-discipline and protect animals. Transcriptomics is a science that studies all transcripts of a specific cell, tissue or organ at a specific stage of growth and development or under a certain physiological condition. Different from the relatively stable genome, transcriptomics will change with the change of growth and development stage, physiological state and external environment. Therefore, we can study the mechanism of biological growth and development, stress physiology, disease resistance and immunity through transcriptome analysis. The behavioral research of Apostichopus japonicus is of great significance for improving the production management mode, improving the breeding mode, improving the harvesting efficiency, designing the harvesting facilities and investigating. The transcriptomics of Apostichopus japonicus is more like a key, which can better help us open the door of behavioral research, help us better understand the causes and regulatory mechanism of Apostichopus japonicus behavior, and make more efficient use of Apostichopus japonicus behavior for production and breeding. In this paper, the research status on different behaviors of Apostichopus japonicus, such as movement, feeding, clustering, stress, reproduction, competition and hibernation, was reviewed, and the transcriptomics related to the behavior of Apostichopus japonicus was summarized, so as to provide reference for the behavioral ecology and transcriptomics of Apostichopus japonicus in the future.

Goldfish industry in Tianjin, which is one of the three goldfish headstreams in China, with more than three hundred years of history is an important part of Tianjin aquarium industry. Through an extensive investigation into practitioners of goldfish industry, the study collected the industry data and historical materials to describe the development history and development status of goldfish industry in Tianjin. The results indicated a fact of scale declining of goldfish keeping, the decline or even lose of traditionally dominant varieties and the aging of practitioner in goldfish industry of Tianjin. Finally, the method of PEST was used to analyse the factors that affected the development of goldfish industry in Tianjin to find corresponding strategies. The study aimed to provide ideas for the healthy development of the goldfish industry in Tianjin and references for the formulation of the industrial policies.

In order to explore the genetic evolution of Escherichia in different countries and regions, and to clarify the genetic relationship of E. in different countries and regions, bioinformatics methods were used to analyze and compare the 16S rRNA genes obtained from laboratory isolation and NCBI, and construct molecular evolutionary tree after pruning. The results showed that there was a certain genetic relationship between the strains of E. coli, which may be the relationship between ancestors and offspring, there were varying degrees of variation, and the ability of variation among different strains had different levels. The transformation ability of E. coli and E. fergusonii was the strongest, and the two strains evolved from the same original strain in a very early period. E. albertii strains coexisted in the same region with highly conservative strains, and cross-evolution existed among Asia, Oceania and Antarctica. E. marmotae and E. vulneris strains originated in the Asian continent, and E. marmotae strains from China were clustered into three groups. E. Marmota had two branches of cross evolution in China, India and South Korea. E. marmota was closely related to E. vulneris. The results provide reference for elucidating the molecular epidemiology of Escherichia.

In order to solve the problem of low basic rural public service and insufficient supply, we should deepen the research on the mechanism of basic public service input in urban and rural areas to provide a guarantee for the “short board” in rural areas. With red beetroot( Beta vulgaris L. var. rapacea Koch.)cultivar ‘BR07-5’ as the experimental material, effect of different sowing dates, different covering soil thickness after sowing, planting and sowing patterns and different harvest dates on growth and yield of red beetroot were studied in Tianjin area. The results showed that individual root weight and yield of red beetroot sowed in late summer and early autumn were significantly decreased as postponing sowing date, but fleshy root texture was firmer when red beetroot was sowed too early, so it was confirmed that the appropriate sowing date was August fifth. Seedling emergence time of treatment of covering soil 0.5 cm after sowing was significantly shorter than those of treatments of 1.5 cm and 2.5 cm and seedling emergence rate of treatment of covering soil 0.5 cm after sowing was significantly higher than those of other two treatments. The plant height of transplanting seedling treatment was significantly lower than that of direct sowing treatment, while leaf number and leaf weight were greater than that of direct sowing treatment, transverse diameter and vertical diameter of fleshy root, individual root weight and yield were all lower than that of direct sowing treatment. While thinning seedling we could remedy missing seedlings and ensure higher yield through transplanting seedling. Individual root weight, yield and texture of red beetroot harvested in November seventh were better.

In this study, a dominant strain CTJ206 was isolated from diseased Cyprinus carpio var. koi. The experimental results of artificial infection showed that the strain CTJ206 was the pathogen of Cyprinus carpio var. koi. By analyzing the physiological and biochemical characteristics of strain CTJ206 and the conserved gene sequences of 16S rDNA, gyrB gene and rpoD gene, we identified the strain CTJ206 as Aeromonas veronii. The healthy Cyprinus carpio var. koi were artificially infected with Aeromonas veronii, and the results showed that the LD 50 of the strain to Cyprinus carpio var. koi was 1.77 × 10 6 CFU/g. We analyzed the common virulence genes of pathogenic bacteria, and the results showed that the strain carried 8 virulence genes of ahyB、ascV、gcaT、act、lip、exu、aerA and fla. Histopathological changes in Cyprinus carpio var. koi caused by Aeromonas veronii infection caused necrosis of liver tissue cells and hemosiderin deposition in spleen tissue. The drug susceptibility test results showed that the strain was highly sensitive to azithromycin, tetracycline, doxycycline, norfloxacin, ciprofloxacin, enrofloxacin and other drugs, and has developed resistance to erythromycin, vancomycin, etc. The results of this study can provide a reference for the prevention and treatment of bacterial diseases of Cyprinus carpio var. koi.

In order to reduce the field irrigation time, design reasonable border field specifications, and improve irrigation efficiency, this study adopted the method of combining the field border double-sided irrigation experiment and WinSRFR5.1 model analysis to simulate the flow process and irrigation quality of double-sided surface irrigation. Taking the water flow confluence as the boundary, the field border double-sided irrigation test was divided into two irrigation tests with different slopes, and combined with the WinSRFR5.1 model to calibrate the soil infiltration parameters(infiltration coefficient K and infiltration index a)and simulate the irrigation quality contour map, and then compared and analyzed the simulation results. The results showed that field border double-sided irrigation was feasible. Compared with border width, border length changes had a greater impact on irrigation efficiency and irrigation uniformity. Combining with the status quo of border fields in Wuqing district, Tianjin city, the border length and width range of bilateral irrigation border fields were obtained: single width flow rate is 2.96 L/(s·m), border width is 2.0-11.5 m, border length is 49-165 m, and irrigation efficiency and irrigation uniformity are both above 90%.

Research on optimization of cider clarification process by response surface methodology

With cider as the research object, through single factor and response surface test design, this experiment has studied the effects of different clarifiers(bentonite, gelatin, PVPP and egg white powder), clarification temperature and clarification time on the clarification effect of apple cider respectively. The alcohol content, total acid, total sugar, light transmittance and sensory scores before and after the wine clarification process were used to optimize the cider clarification process. The results showed that bentonite had the best clarification effect on cider, with the highest sensory score. The best clarification conditions were: bentonite addition amount 1.02 g/L, clarification temperature 9.6 ℃, clarification time 42.2 h. Under these conditions, the cider was transparent, the light rate was 99.7%. Compared with the unclarified cider, the light transmittance was increased by 24.9%. Compared with the original wine, the total sugar, total acid and alcohol content of the clarified cider were basically unchanged.

To understand the species of oysters in the coastal area of Tianjin, a total of 127 wild oysters were collected from three sampling sites, including the Duliujianhe river estuary, Beijiang Power Plant Breakwater and Dongjiang Port. First, morphological indexes were measured, and then cytochrome oxidase I(COI)gene sequence was analyzed. The oyster species were further identified. The results showed that 9 haplotypes were obtained from 16 COI gene sequences. The Haplotype diversity( h)and Nucleotide diversity( π)were 0.767 and 0.001 73 respectively. Cluster analysis results showed that the oyster sequences obtained in this study first clustered into a branch with Crassostrea gigas, and then clustered into a branch with Crassostrea angulata. Based on the results of morphological observation and molecular identification, the oyster samples collected from the three locations along the coast of Tianjin were all Crassostrea gigas, indicating that the composition of oyster species in Tianjin sea area was relatively simple. The Talonostrea talonata and Crassostrea ariakensis as previously reported were not found in this study, and further sampling sites are needed to obtain more comprehensive conclusions. The results of this study are of great significance for understanding the status of existing oyster resources and protecting species diversity in Tianjin.

Improvement effect of saline-alkali soil by the cultivation of Stropharia rugosoannulata

The aim of the article is to investigate the effects on pH, total salt content, organic matter, available potassium and ammonium nitrogen content of the soil after planting Stropharia rugosoannulata in it, and study the improvement on saline-alkali soil by planting S. rugosoannulata. The results showed that compared with the control group CK, the pH value and total salt content of experimental group A in experimental site I and experimental site II were reduced, and the pH of experimental group A in experimental site I was reduced by 2.94% compared with control CK. The total salt content was reduced by 42.36%. Compared with the control CK, the pH of experimental group A in Experimental Site II was reduced by 2.46%, and the total salt content was reduced by 3.08%. Compared with the control group CK, the contents of organic matter, available potassium and ammonium nitrogen in experimental group A in experimental site I and experimental site II increased significantly. Among them, the available potassium in experimental site I increased by 6.17% and ammonium nitrogen increased by 11.85% ; The soil sample available potassium in Experimental Site II increased by 76.77% and ammonium nitrogen increased by 164.39%. It shows that the Stropharia rugosoannulata has a positive effect on improving the physical and chemical properties of saline-alkali soil.
