Identification and pathogenicity analysis of Aeromonas veronii from Cyprinus carpio var. koi

Abstract: In this study, a dominant strain CTJ206 was isolated from diseased Cyprinus carpio var. koi. The experimental results of artificial infection showed that the strain CTJ206 was the pathogen of Cyprinus carpio var. koi. By analyzing the physiological and biochemical characteristics of strain CTJ206 and the conserved gene sequences of 16S rDNA, gyrB gene and rpoD gene, we identified the strain CTJ206 as Aeromonas veronii. The healthy Cyprinus carpio var. koi were artificially infected with Aeromonas veronii, and the results showed that the LD50 of the strain to Cyprinus carpio var. koi was 1.77 × 106 CFU/g. We analyzed the common virulence genes of pathogenic bacteria, and the results showed that the strain carried 8 virulence genes of ahyB、ascV、gcaT、act、lip、exu、aerA and fla. Histopathological changes in Cyprinus carpio var. koi caused by Aeromonas veronii infection caused necrosis of liver tissue cells and hemosiderin deposition in spleen tissue. The drug susceptibility test results showed that the strain was highly sensitive to azithromycin, tetracycline, doxycycline, norfloxacin, ciprofloxacin, enrofloxacin and other drugs, and has developed resistance to erythromycin, vancomycin, etc. The results of this study can provide a reference for the prevention and treatment of bacterial diseases of Cyprinus carpio var. koi.
