The new record of Trichopria drosophilae from Tianjin, China

Trichopria drosophilae(Perkins, 1910)was first recorded in Tianjin, China. Identification of T. drosophilae based on morphological characters and molecular data was provided first in China. Detailed morphological description and adult photographs of the species were given, with a brief cladistic analysis on some Trichopria species with available mitochondrial gene COI sequences.

Analysis of main nutrients from Mercenaria mercenaria in salt field

In this paper, the basic nutrients, amino acids, fatty acids and minerals of Mercenaria mercenaria were determined and analyzed. The results showed that the moisture content of the M. mercenaria was 86.37%, and the content of protein, fat and ash in the dry base were 66.92%, 6.21% and 16.96% respectively. A total of 21 fatty acids were detected, including 10 saturated fatty acids(SFA), 4 monounsatura-ted fatty acids(MUFA)and 7 polyunsaturated fatty acids(PUFA). The content of unsaturated fatty acid was 52.88% of the total fatty acids. The content of EPA and DHA was 65.10% of the PUFA, 27.18% of the total fatty acids. 16 amino acids were detected. The content of delicious amino acid was about 40.82% of the total amino acids. The EAA/TAA was 35.79% and the EAA/NEAA was 55.7%. The contents of Na, Ca, K, Mg were rich. In summary, the nutritional value of the M. mercenaria was high.

According to the requirements of engineering education certification and “First-class Courses”, this paper analyzed the problems existing in classroom teaching by taking Hydrology Principle course as an example, combining with the actual situation of talent cultivation of hydrology and water resources engineering major. The curriculum teaching system was constructed by teaching reform from the aspects of teaching content selection, diversified teaching methods, multi-dimensional integration of ideological and political elements, and diversified assessment methods. Through class performance, section tests, assignment, experiment and examination, the achievement of each student’s course goal was evaluated comprehensively. With questionnaire survey the learning effect of students was aquired. The teaching results showed that the above reforms could effectively improve students’ interest in learning and deepen their understanding of knowledge. The course objectives were well achieved, and the teaching process could be continuously improved based on the evaluation results.

The extraction of purple cabbage pigment and the development of lipstick

In order to develop the natural purple cabbage pigment lipstick product, extracted purple cabbage pigment was used as the main pigment raw material. Single factor and orthogonal experiments were used to determine the optimal extraction conditions for extracting purple cabbage pigment. The optimal condition was that extraction temperature was 80 ℃; concentration of hydrochloric acid was 0.35 mol/L; extraction time was 60 min. The changes of pigment color of purple cabbage under different pH values were studied. The optimal formula of lipstick determined by single factor test was 41% purple cabbage pigment, 27.2% olive oil, 2.8% candela wax, 27.2% white beeswax, and 1.8% emulsifier. The purple cabbage pigment lipsticks prepared with smooth texture, good hardness, safety, health and beautiful color.

In this study, leaf, petiole and hypocotyl of Gegu radish were used as explants to induce loose callus,to explore the effect of different explants and different hormone concentrations on the induction of loose callus. The results showed that different explants all could induce callus, but only cotyledon and petiole could induce loose callus, and the loose callus induced by cotyledon grew faster, and the callus induction rate was(26.88±1.44)%. Cotyledon was the most suitable explant for inducing loose callus. The optimal medium for induction of loose callus was MS+0.25 mg/L6-BA+2.0 mg/L 2,4-D. The induction rate of loose callus was(95.83±2.20)%, and the browning rate was only(0.83±0.83)%.

In order to investigate the discharge and treatment of tailwater from factory aquaculture in Tianjin, ten representative aquaculture enterprises were selected to investigate the tailwater quality. 57 water samples were collected from May 2019 to March 2021 and according to the “Sewage Requirements of Mariculture Tailwater”(2007 edition)the inorganic nitrogen, COD, suspended solids, active phosphate, pH and salinity were determined respectively. The results showed that inorganic nitrogen, COD, suspended solids and active phosphate all exceeded the standard, and the exceeding rates were 22.81%, 26.32% 84.21%, and 85.96%, respectively. Therefore, at present, the overall water quality of industrial aquaculture tailwater in Tianjin was not good enough and the existing tailwater treatment efficiency still needs to be further improved; the relevant standards in “Sewage Requirements of Mariculture Tailwater”(2007 Edition)are too strict and are not applicable for the tailwater from industrial aquaculture.

Comparison of morphological structures of leaves and physiological characteristics of two Jujube varieties

Annual grafted seedlings(rootstocks were sour jujubes)of two jujube varieties, ‘Yueguang’ and ‘Fucuimi’, were used as the test material for the study. The morphological structures of young leaves of two jujube varieties were determined by microscopic micrometer, and their leaf physiological indexes and photosynthetic pigment contents were measured to explore the suitability of two jujube varieties in the Tianjin area and provide a reference for its introduction and cultivation in the Tianjin area. The results showed that no stomata were found on the upper epidermis of the leaves of either jujube variety, and stomata were mainly distributed on the lower epidermis. The thickness of young jujube leaves was 191.01 μm, the thickness of upper and lower epidermal cells were 28.69 and 22.29 μm respectively, the thickness of lower cuticle of leaves was 3.28 μm, the thickness of xylem of leaf veins was 107.99 μm, and all these indexes were significantly greater than those of ‘Yueguang’. Comprehensive analysis showed that compared with ‘Yueguang’, the leaf structure of young ‘Fucuimi’ jujube trees had enhanced water retention, water transport and photosynthetic capacity, making ‘Fucuimi’ more suitable for introduction and promotion in Tianjin area.

Effect of enzymatic hydrolysis on soluble dietary fiber and antioxidant activity in old asparagus stalks

The large amount of discarded old stems generated during the process of asparagus harvesting and processing contains rich dietary fiber and active substances, which have certain antioxidant effects and can be used to prepare asparagus old stems with antioxidant activity. Using asparagus stems rich in dietary fiber as raw materials, the content of soluble dietary fiber was increased through cellulase hydrolysis, thereby enhancing its antioxidant activity. By measuring the antioxidant activity and soluble dietary fiber content of asparagus stem liquid before and after enzymatic hydrolysis, single factor and orthogonal optimization experiments were conducted to study the enzymatic hydrolysis time, enzyme addition amount, enzymatic hydrolysis temperature, and determine the optimal enzymatic hydrolysis conditions.The results showed that under the conditions of cellulase addition of 1.0%, enzymolysis time of 1.75 h, and enzymolysis temperature of 50 ℃, the content of soluble dietary fiber increased by 2.9 times, the scavenging capacity of hydroxyl radical increased by 1.69 times, and the content of active substances such as polysaccharides, flavonoids, and saponins increased by 1.39, 1.56, and 1.84 times respectively compared with non enzymolysis.This study can provide a certain theoretical basis for the reuse of old asparagus stems.

In order to understand the spatial variation characteristics of leaf light response during the grain-filling stage of summer maize in North China, taking summer maize in North China as the research object, the light response curves of different leaf positions and different parts of the leaf during the grain filling period of summer maize were observed through field experiments. The results showed that there were significant differences in the response of photosynthetic characteristics to photosynthetically active radiation in different leaf positions and different parts of leaves during the grain filling stage of summer maize, and the photosynthetic parameters obtained by the light response model were significantly different in the spatial distribution of plants. The research results can provide reference for improving the growth and development environment of summer maize during grain-filling period in North China.

In order to understand the microdevelopment characteristics of intestine and liver of juvenile Cynoglossus semilaevis at different stages, paraffin sections, hematoxylin and eosin staining, microscopic observation and other methods were used to compare the intestinal microstructural characteristics of C. semilaevis with an average body weight of 2 g(W 2), 26 g(W 26), 59 g(W 59), and 228 g(W 228). The number of mucosal folds, the height of mucosal folds, the thickness of muscle layer, the relative density of mucinous cells and the height of monolayer columnar epithelium were measured. The results showed that the number of mucosal folds, the height of mucosal folds, the length of striatum margin and the single columnar epithelium of the four groups showed a consistent downward trend along the intestinal tract from front to back. Mucinous cells were distributed everywhere in the intestine, and their density gradually increased from front to back, and the change pattern of the four groups was consistent. Except for the number of mucosal folds and the density of mucinous cells, the other four morphological indexes showed a downward trend in the anterior part of the intestine, but gradually increased with ontogenetic development. The striatum border and the simple columnar epithelium of the midgut and hindgut decreased first and then increased in the four groups. The thickness of hindgut muscle layer decreased obviously, showing certain regularity. The liver was well developed in the four developmental stages, and the hepatocytes were polygonal. In conclusion, the intestinal and hepatic structures of C. semilaevis were gradually refined as the individuals developed, and in the W 228 group the intestinal and hepatic structures were well developed.

Investigation on the immune effect of Vibrio vulnificus inactivated vaccine in fish

In order to study the immune effect of inactivated Vibrio vulnificus vaccine, this study prepared Vibrio vulnificus microsphere vaccine by inactivating the Vibrio vulnificus strain EPL0201 stored in the laboratory with 0.3% formaldehyde and wrapping the inactivated bacteria solution with sodium alginate. Healthy pearl gentian grouper was immunized by oral route, and was injected intraperitoneally with 1×10 7 cfu/mL Vibrio vulnificus to challenge the pearl gentian grouper. The expression trend of some immune related genes of the pearl gentian grouper after immunization and the immune protection rate after challenge were detected. The results showed that the particle size of Vibrio vulnificus microsphere oral vaccine was(34.55±5.33)μm. The coverage rate was 90.5%. Its safety was good. Quantitative real-time PCR(QRT-PCR)results showed that, compared with the control group, each immune gene had different expression changes successively. They participated in immune response in kidney, spleen, liver and intestine which involved the genes TLR1, TLR2, TLR3, TLR5, and Myd88 in the TLR signaling pathway and inflammatory cytokines IL-1β, IL-12P40 and TNF-α. The expression of immune genes in different tissues was different, and the highest expression level of IL-1β was in spleen, and the highest value was 72.37 times that of the control group. The peak of peak expression of the same immune gene in different organs and tissues of fish was different, and the peak expression of different immune genes in the same tissue was also different, which indicated that the gene expression changes in different tissues were different. The results of challenge experiment showed that the immune protection rate was 61.3%. The results of this study showed that Vibrio vulnificus oral vaccine can induce immune genes to participate in the immune response, and has immune protection effect on pearl Gentian grouper. It also provides experimental data support for the prevention and treatment of diseases caused by Vibrio vulnificus with microsphere oral vaccine.

Clinical safety of canine adipose derived mesenchymal stem cells in vein transplantation

In order to verify the safety of clinical application of canine adipose derived stem cells (ASCs), the fourth generation of canine ASCs were selected in this experiment, and the cell morphology was observed under the microscope, and adipogenesis, osteogenesis and chondrogenesis were induced and differentiated, and cell surface marker proteins were analyzed. The results showed that ASCs grew as spindle-like adherent cells with uniform morphology. ASCs could differentiate into adipocytes, osteocytes and chondrocytes in vitro. Canine ASCs express marker proteins CD29, CD44, CD105 and CD166, but do not express CD34 and CD45, which conforms to the typical characteristics of adipose mesenchymal stem cells. Canine ASCs with 1×10 6/kg vein transplantation was performed at a speed of 10 mL/(kg·h). No adverse reaction was found during the injection. Long term observation for 3 months showed no obvious complications. Compared with that before transplantation, the number of white blood cells, neutrophils, monocytes, eosinophils, red blood cells, hemoglobin and platelets showed no significant difference( P>0.05); Serum biochemical examination showed that there were no significant differences in liver indicators such as albumin, ALT, AKPP>0.05), renal indicators such as creatinine, serum urea, blood phosphorus and blood calcium( P>0.05), pancreatic related indicators such as blood glucose and serum amylase( P>0.05), myocardial injury indicators such as creatine kinase and blood lipid indicators such as cholesterol before and after cell transplantation( P>0.05). The results of this study showed the clinical safety of ASCs in dogs and laid a foundation for cell transplantation therapy.

Effects of insect feces organic fertilizer on physical and chemical properties of tobacco-planting soil and tobacco growth

Pot experiment of flue-cured tobacco ‘Zhong Yan 100’, carried out to explore the effects of earthworm casts fertilizer, white grubs feces fertilizer and black soldier fly feces fertilizer on soil physicochemical properties and tobacco growth, with traditional sesame cake fertilizer as control. The results showed that the contents of available phosphorus and available potassium in soil and the activities of phosphatase, urease and invertase in soil treated with black soldier fly feces fertilizer increased by 65.66%, 23.36%, 12.20%, 24.39% and 23.61%, respectively, compared with those treated with sesame cake fertilizer. The contents of available phosphorus and available potassium and urease activity in soil treated with white grubs feces fertilizer were not significantly different from those treated with sesame cake fertilizer, while the contents of soil organic matter and alkali-hydrolyzable nitrogen and the activities of soil phosphatase and invertase were decreased. The soil nutrient indexes and enzyme activities of earthworm casts fertilizer were significantly lower than those of sesame cake fertilizer. The soil pH of the three kinds of insect feces organic fertilizer were increased, while the soil EC of the white grubs feces fertilizer and the black soldier fly feces fertilizer were increased. The tobacco plant height, maximum leaf area, root vigor and root dry weight of black soldier fly feces fertilizer were higher than those of sesame cake fertilizer, while those of earthworm casts fertilizer and white grubs feces fertilizer were lower than those of sesame cake fertilizer. At the same time, the contents of nicotine and sugar in tobacco leaves were increased by the treatment of black soldier fly feces fertilizer and white grubs feces fertilizer, and the contents of potassium, chlorine and potassium to chlorine ratio in tobacco leaves were in or close to the appropriate range, and the quality was better. In conclusion, among the three kinds of insect feces organic fertilizer treatments, black soldier fly feces fertilizer had the best fertilizer effect, which was superior to traditional sesame cake fertilizer, improved the nutrient content of tobacco soil, enhanced soil enzyme activity, and promoted tobacco growth and quality improvement.

Effects of different control irrigation modes on the main agronomic characters and quality of rice

In order to study the water management mode of rice quality, with rice variety ‘Jinyuan 85’ as the experimental material, four different control irrigation modes were arranged , namely 60%(W1), 70%(W2), 80%(W3)and 90%(W4)of soil saturation water content as the irrigation limit, and the effects of different control irrigation modes on the main agronomic traits and japonica rice quality of rice were analyzed with conventional irrigation(CK)as the control. The results showed that compared with the control, different control irrigation modes had inhibitory effects on rice plant height, stem and leaf dry weight, but could promote the increase of moisture content in rice ears. The quality of rice under different controlled irrigation modes was better than that of conventional irrigation. The amylose content W2 was the highest, which was 0.27% higher than that of CK. Different control irrigation modes reduced irrigation water consumption by 3 990-5 415 m³/hm² compared with conventional irrigation. Through the comprehensive evaluation of various rice quality indicators and the sum of irrigation and rainfall, it is found that controlling soil saturation moisture content of 90% as the limit of irrigation water is the best rice water management mode.

In order to explore the effects of colchicine treatment on the seed germination and seedling growth of broccoli, this experiment took broccoli seeds as materials, soaked with different concentrations of colchicine(0.03%, 0.05%, 0.07%, 0.10%, 0.20%, 0.30%)for different time(12, 24 h), and compared with the seed soaking treatment in distilled water(CK). The effects of colchicine treatment on the seed germination, seedling morphological, physiological and biochemical characteristics of broccoli were studied. The results showed that compared with CK, there was no significant difference in germination rate and germination potential among the treatment groups after soaking for 12 h, and the vitality index of 0.20% and 0.30% colchicine trearment groups increased significantly. After soaking for 24 h, all germination indexes decreased in all treatment groups. 0.10%-0.30% colchicine treatment increased the fresh weight and stem thickness of broccoli seedlings, decreased plant height, leaf length and width, increased the antioxidant enzyme(SOD, CAT)activities, osmoregulatory substances and glucosinolates content, while decreased the malondialdehyde content. According to the analysis of membership functions of the measurement indicators of broccoli, the average membership function values of 0.30% colchicine soaking for 12 and 24 h ranked first. In a comprehensive analysis, 0.30% colchicine soaking for 12 h was considered as the best treatment combination to promote seed germination and seedling growth of broccoli.

In this paper, the factors affecting the growth of Dendrobium officinale tissue culture seedlings were studied in order to optimize the rapid propagation system of Dendrobium officinale tissue culture. The results showed that low salt medium 1/2 MS was more suitable for the growth of Dendrobium tissue culture seedlings; the effect of adding 0.2 mg/L NAA on the growth of Dendrobium officinale tissue culture seedlings was significantly higher than other auxin; when sucrose concentration was 25 g/L and inorganic salt concentration was 1/2 MS, the osmotic pressure environment of culture medium was more suitable for the growth of D endrobium officinale tissue culture seedlings; organic additives LH(hydrolyzed milk protein)and CH(hydrolyzed casein)could promote the growth of Dendrobium officinale tissue culture seedlings, among which 0.1 g/L hydrolyzed milk protein was the most suitable for the growth of Dendrobium officinale tissue culture seedlings, where 0.8 cm tissue culture seedlings, after 60 days of culture, reached the average plant height 2.26 cm, stem diameter 2.73 mm, rooting number 2.45, tillering amount 3.51, and the plant grew vigorously with the leaves large and dark green. This study lays a foundation for the establishment of efficient tissue culture and rapid propagation of D endrobium officinale.

The effects of different concentrations of microbial inoculants on the growth, quality and soil properties of the‘Qixing’ fruit radish were studied by field experiment, in order to provide a reference for the most suitable amount of microbial inoculants for the cultivation of radish in greenhouses. The results showed that plant height, stem diameter, leaf area and leaf color were affected in different degrees, but the difference was not significant. The total output of radish under T4 treatment was the highest, and the economic benefit reached 230 900 yuan/hm 2. For the quality of radish in facilities, the vitamin C content of T3 treatment was the highest, which was increased by 17.9% and 11.0% respectively compared with T1 treatment without inoculant and T7 treatment, followed by T5 treatment. The total acidity of T6 treatment was the lowest, and the sugar-acid ratio was the highest(8.65), which was significantly increased by 19.24% compared with T1 treatment without inoculant, but had no significant difference compared with T7 treatment. The reduction of nitrate content was most significant in T2 treatment. In rhizosphere soil, urease activity was the highest in T3 treatment, followed by T5 treatment, alkaline phosphatase activity was the highest in T1 treatment, followed by T3 treatment. In the non-rhizosphere soil, the activities of urease and alkaline phosphatase in T4 treatment decreased the most. In the surface soil, the residual content of available phosphorus was the lowest in T3 treatment, followed by T4 treatment, the residual content of nitrate was the lowest in T2, T3 and T4 treatment, and the contents of organic matter and available potassium were the highest in T5 treatment. The results showed that the application of appropriate concentration of microbial agents could promote the growth of radish, improve the yield and quality of radish, improve the soil properties of facilities, and reduce environmental pollution. Overall, T4 treatment(360 kg/hm 2)had the best effect on the growth and soil properties of facilities radish, followed by T3 treatment(180 kg/hm 2).

Effects of biogas slurry application on rice growth and soil nutrients

The effects of different biogas slurry dosages(0, 75, 150, 225, 300 m 3/hm 2)and chemical fertilizer(conventional and halved)on rice growth and soil nutrients were studied through field experiments for two consecutive years. The results showed that the rice yield was the highest when the amount of biogas slurry was 150 m 3/hm 2+halved chemical fertilizer, and compared with the conventional pure chemical fertilizer treatment, the rice yield increased by 2.13% and 5.79% in the first and second years respectively. The biomass of rice stems and leaves was the highest when the amount of biogas slurry was 150 m 3/hm 2 + conventional chemical fertilizer. When the amount of biogas slurry exceeded 225 m 3/hm 2, the yield, plant height and biomass of rice stem and leaves decreased. The application of biogas slurry increased the nutrient content of soil, which increased with the increase of application amount. At the same application amount of chemical fertilizer, the content of soil organic matter increased by 8.93%~30.04%, the content of alkali hydrolyzable nitrogen increased by 4.36%~39.90%, the content of available phosphorus increased by 14.86%~39.89%, and the content of available potassium increased by 9.17%~41.25%. Biogas slurry application had no significant effect on soil pH but increased soil EC value; however, soil EC value did not increase in the second year of the same treatment. In conclusion, in this experiment, the amount of biogas slurry 150 m 3/hm 2 and halved conventional chemical fertilizer is is the best fertilization treatment.

In this study, a strain ZYRH-19 producing extracellular polysaccharide was isolated from soil. It was classified and identified from the perspectives of morphology, physiology, biochemistry, and 16S rDNA gene sequence, and the extracellular polysaccharide synthesized by strain ZYRH-19 was analyzed and identified. The results showed that ZYRH-19 was a strain of Sphingomonas sp., and its homology with Sphingomonas sp. ATCC 31555 was 99.93%. The extracellular polysaccharide synthesized by strain ZYRH-19 was welan gum with weight average molecular weight of 5.3×10 6 u. The welan gum solution synthesized by ZYRH-19 had good dilution shear property at room temperature. The viscosity of the product solution could reach 1 444.1 mPa·s at the 6 r/min shear rate, and its gel strength was 22.6 g. It had good temperature resistance at 160 ℃. The results showed that the performance of welan gum synthesized by strain ZYRH-19 is very good, which provides a new strain resource for the industrial production of welan gum.

In order to explore the adsorption performance of carbon aerogels modified by NaOH/H 3PO 4 for methylene blue, carbon aerogels was modified by chemical activation method. The effects of pH value of methylene blue solution, initial concentration of solution, gel adsorption time and gel dosage on the adsorption effect of carbon aerogels modified by NaOH/H 3PO 4 were studied through batch experiments. The experimental results showed that the adsorption capacity of modified carbon aerogels for methylene blue decreased with the increase of pH, and the adsorption effect of modified carbon aerogels for methylene blue was the best when pH=8. The adsorption capacity of modified carbon aerogels could be improved by prolonging the gel adsorption time, increasing the initial concentration of solution and increasing the gel input. Under the conditions of dye solution pH=8, initial concentration 50 mg/L, adsorption time 12 h and gel input 5 mg/mL, the adsorption capacity of NaOH modified gel could reach 7.51 mg/g, and that of H 3PO 4 modified gel can reach 4.29 mg/g, both of which were higher than that of unmodified carbon aerogels, it indicates that acid-base modification can improve the adsorption performance of watermelon peel-based carbon aerogels.

These countermeasures should be implemented to help ex-servicemen better serve rural revitalization including perfecting the career development path of ex-servicemen, opening up channels for ex-servicemen to serve rural governance, optimizing the employment and entrepreneurship service system for ex-servicemen, reinforcing training and education to empower ex-servicemen, and strengthening publicity and guidance.

The effects of nano-silver and 1-MCP treatment on vase life and flower diameter, relative fresh weight(RFW)and vase solution uptake(VSU), relative chlorophyll content and malondialdehyde(MDA)content, relative conductivity and antioxidant capacity of cut rose ‘Carola’ were studied. The results showed that compared with the control, the single and combined treatment of nano-silver and 1-MCP could significantly improve the reducing power, increase the activities of ascorbate peroxidase(APX), superoxide dismutase(SOD)and catalase(CAT), decrease the content of malondialdehyde(MDA)in petals and the relative conductivity in leaves, and inhibit the decrease of chlorophyll content. Compared with nano-silver and 1-MCP alone, the combined treatment of nano-silver and 1-MCP was more effective in prolonging the vase life, increasing the diameter of cut flowers, promoting the water absorption of cut flowers, maintaining the plasma membrane stability of cut flowers and improving the antioxidant capacity of cut flowers. In conclusion, the combination of nano-silver and 1-MCP can prolong the vase life of cut rose by maintaining the water balance and plasma membrane stability, improving the antioxidant capacity and reducing the membrane lipid peroxidation level of cut flowers.

In order to study the factors that affect the rapid growth of Dendrobium nobile tissue culture seedlings, this experiment took Dendrobium nobile tissue culture seedlings as the experimental material, and set up an orthogonal test at the three levels of auxin NAA, hydrolyzed milk protein, organic components of MS medium, and activated carbon to observe the growth performance of Dendrobium nobile tissue culture seedlings, so as to determine the best medium combination. The results showed that the combination of 0.10 g/L activated carbon+0.50 mg/L NAA+0.05 g/L hydrolyzed milk protein+1 fold organic component had the best effect on the growth of tissue culture seedlings; and NAA can significantly increase the growth height and coarseness of tissue culture seedlings. This study lays a foundation for the study of tissue culture and rapid propagation of Dendrobium nobile.

Using the organic fertilizer made from spent mushroom substrates produced after the cultivation of Pleurotus ostreatus with corn straw as the base fertilizer, the indoor pot experiment was carried out. The results showed that the application of spent mushroom substrates fertilizer had a certain promoting effect on the growth and development of maize seedlings stage. Compared with the control group, the plant height of maize seedlings cultivated with 12 g spent mushroom substrates fertilizer per kilogram of soil was increased by 18%, the aboveground fresh weight was increased by 23.27%, the aboveground dry weight increased by 23.89%, and the chlorophyll content and photosynthetic rate also increased significantly. The further test results showed that the application of spent Pleurotus ostreatus substrates fertilizer increased the content of soil organic matter, soil conductivity, soil cation exchange capacity, soil available nutrients and soil total nutrients, and could decrease the pH value of soil.

In order to improve the diagnostic accuracy of cat renal failure and optimize the treatment scheme of cat renal failure, nine cats with renal failure were collected from a pet hospital of Tianjin in 2021. According to the medical history, clinical examination and laboratory test, the causes of renal failure were comprehensively analyzed, and the combination of infusion and diet was used to treat cat renal failure. The results showed that the etiology of cat renal failure was diverse, and the incidence type was related to age. After the treatment, 9 cats with renal failure were cured and discharged. This paper provides a reference for the diagnosis and treatment of cat renal failure.

Peanut is one of the most common and important food allergens. In this paper, peanut oil was degreased by petroleum ether and purified by ammonium sulfate salting out, dialysis and Sephadex G-150 dextran gel column. The molecular weight of peanut protein was determined by SDS-PAGE gel electrophoresis: 15, 20, 35 and 60 ku. Western-Blotting immunoblot test showed that peanut allergy was around 63.5 ku. 31 effective protein peptides were obtained by mass spectrometry sequencing. Among them, 5 peptides had higher number of spectra, namely, KLEYDPRCVYDTGAT, AFNAEFNEIRRVLLEE, DNVIDQIEKQAK, PYSPSQDPDRRDPY and QDPYSPSQDPDR, which accounted for 49.6% of the total number of spectra. Based on this, it is inferred that those peptides are the main peptide allergic sequences of peanut sensitizing proteins Ara h 1 and Ara h 2. The protein peptides are successfully used to infer the corresponding protein types, which lays a foundation for further clarifying the sensitization mechanism of peanut allergens. Since the conformational structure of peanut allergic protein is complex, further research work needs to be carried out by other means

As a new type of high-density aquaculture method, recirculating aquaculture system has the advantages of controllable aquaculture water environment, stable system operation, safe and reliable technology, and high water saving and land saving. In this paper, the operation effect of recirculating aquaculture system was analyzed by taking the cultivation of Cynoglossus semilaevis in an enterprise in Binhai New Area of Tianjin as an example. The results showed that the culture density of Cynoglossus semilaevis was(20-25)kg/m 2, and the initial average mass was 5 g. After 407 days of culture in recirculating aquaculture system, the final average mass was 1 079 g, and the survival rate was 29.1%. The average removal rates of ammonia nitrogen, nitrite, vibrio and bacteria were 64%, 58% and 99%, respectively. The pH and salinity were 7.0-8.5 and 18-35, respectively. The results of economic analysis show that the system is feasible and economical, and it is suitable to be popularized in coastal areas.

In order to keep the strong flavor of the river crab enzymolysis liquid(RCEL)in the process of storage, transportation, processing and utilization and avoid its volatilization, RCEL was prepared by the complex coacervation method. With gelatin alginate solution and gelatin sodium carboxymethyl cellulose(CMC-Na)solution as wall materials respectively, RCEL was used as core material to study the influence of pH value, wall material ratio and wall core ratio on the encapsulation effect of microencapsulated emulsion in the condition of complex coagulation. The experimental results showed that the optimum process of selecting gelatin sodium alginate as wall material was as follows: adjusting the pH of the solution to 4.0, the ratio of gelatin to sodium alginate to 2∶1 and the ratio of wall to core to 1∶3; when gelatin CMC Na was selected as wall material: pH 3.4, Gelatin: CMC-Na 2∶1, and wall core ratio 1∶2. The RCEL microcapsules obtained under those two conditions were round, spherical and numerous, and the individuals were homogeneous. When the emulsion was reheated, it could smell strong crab. This research will widen the application field of crab.

Seawater factory farming is a modern high-tech fishery with the advantage of water and energy saving. However, the diseases of the cultured fish happened frequently for the high density and intensive farming model and has caused great economic loss to the culture enterprises. Vaccination is an effective way to control the disease of cultured fish. Compared with injection and soaking, oral vaccine has the advantages of convenient inoculation, no injury to the body and good compliance of the receptor, which has become a hot spot of research. Sodium alginate is a natural macromolecule polysaccharide from the ocean. It is nontoxic, biocompatible and degradable. The degradation products can be fully absorbed by the organism. The use of sodium alginate as the carrier material to prepare the microsphere vaccine can increase the viscosity and prolong the release time of the drug. Sodium alginate is feasible to prepare the oral vaccine for fishing as a carrier for its low price, uncomplicated technology and convenient source. The study developed a new method to generate fish oral vaccine of Edwardsiella tarda by emulsfication with sodium alginate as the carrier. The results showed that the optimum preparation process of oral vaccine was 3%( m/ v)sodium alginate solution, 4%( m/v)calcium chloride solution, 1∶2 ratios of water and oil, 1% Span80 emulgator, 1 200 r/min stirring rate. The embedding rate was 91.04%. The mean grain size of the microballon was(16.51±11.87)µm. It is necessary to prepare the microsphere vaccine suitable for the intestinal targeted drug delivery, and the artificial simulated gastric and intestinal juice were used to analyze the tolerance of vaccine microspheres when treated by them in vitro. The results showed that the releasing rate of the microballon were 0.8% and 98.1% after dealing with simulated gastric and intestinal juice for 16 hours, respectively. Escherichia coli expressing green fluorescent protein emulsficated with sodium alginate was orally administrated to Cynoglossus semilaevis Gunther to detect the tolerance of vaccine microspheres in gastric and intestinal juice in vivo. It showed that the microspheres do not dilate or disintegrate in the acid stomach, or disintegrate in the neutral environment of the intestine. The above results indicated that the vaccine can endure the acid environment of the stomach, reach the hindgut of the fish and be absorbed by the villus, which is suitable for drug targeting. The results of the immunprotection experiment showed that the highest serum titer was 1∶322 after oral vaccinatione for 14 days, the highest leukocyte phagocytosis rate was 27.06% after oral vaccination for 14 days, and the highest phagocytosis index was 1.65 after oral vaccination for 14 days. All fishes were challenged with Edwardsiella tarda after oral vaccination for 28 days, the mortality rate of control group and vaccine group were 58.82% and 13.33% respectively, and the immune protective rate was 77.33%. The present study proposed a new fish oral vaccine preparation technology. The oral vaccine could activate the fish immune response and prevent fish disease.

In order to elucidate the species name and biological characteristics of an isolate from a chicken neck infection, isolation and purification, smear staining microscopy, biochemical test, drug sensitivity test and 16S rRNA gene sequence analysis were carried out. The results showed that the morphological characteristics, biochemical characteristics and 16S rRNA gene sequence of this isolate were consistent with those of Enterococcus faecalis, so the isolate was identified as Enterococcus faecalis. This Enterococcus faecalis was resistant to tetracycline, clindamycin, cotrimoxazole, polymyxin B and vancomycin, moderately sensitive to ciprofloxacin and norfloxacin, and sensitive to benzocillin, erythromycin, chloramphenicol, minocycline etc. The above findings may provide a clinical reference for the prevention and control of Enterococcus faecalis infections.

In response to the low efficiency of manual picking in strawberry planting bases, this paper designs a differential motion walking control system for strawberry picking robots. With focuses on the design requirements of strawberry picking robots, this studyhas conducted an in-depth research and analysis on the kinematics of the mobile chassis. Subsequently, the hardware and software parts of the walking control system for the strawberry picking robot was designed. In terms of hardware, we have selected a main controller with a 32-bit ARM microcontroller as the core, while integrating key components such as power circuit, ultrasonic distance measurement unit and motor drive module. In terms of software, we adopted the CANOPEN protocol, ported it to the microcontroller, and debugged it on the basis of the UCOSII real-time operating system. In addition, we have designed a PID control strategy to provide support for precise control of robots. This comprehensive solution greatly improved the efficiency of strawberry picking and provided strong support for the development of agricultural robotics. With the control system of the strawberry picking robot proposed in this article, more widespread robot application is expected to achieve in the process of agricultural modernization, so as to further promote the development and upgrading of the agricultural industry.

Effects of betaine on seed germination and seedling growth of wheat under salt stress

In order to improve the germination and growth ability of wheat in saline soil, the germination experiment of wheat seeds treated with betaine was carried out under different concentration of NaCl solution, and the effects of betaine on seed germination and seedling growth of wheat under salt stress were studied. The results showed that germination potential, vigor index, fresh and dry weight of seedlings and seedling height of wheat increased significantly treated with 0.05% and 0.10% betaine at 40 mmol/L and 80 mmol/L NaCl, Soaking seeds with 0.05% and 0.10% betaine could significantly increase the root length under 80 mmol/L NaCl, but soaking seeds with 0.05% betaine couldn’t significantly increase the root length under 40 mmol/L NaCl. Seed soaked with 0.05% and 0.10% betaine could improve the germination rate and germination index of wheat seeds, but there was no significant difference compared with the control. Seed soaked with 0.05% and 0.10% betaine could enhance the seedling growth of wheat by increasing seed vigor.

Effects of different phosphorus levels combined with organic materials on the morphology of inorganic phosphorus in fluvo-aquic soil

In this paper, a pot experiment was conducted to plant winter wheat on four phosphorus levels of aquic soil combined with rape green fertilizer, organic fertilizer and corn straw. The contents of inorganic phosphorus components in soil before planting and after harvest of winter wheat were determined by Gu Yichu-Jiang and Baifan method, and the effects of organic materials on the transformation of inorganic phosphorus components in aquic soil were studied. The results showed that the content of inorganic phosphorus increased significantly with the increase of phosphorus application rate, the content of inorganic phosphorus in organic materials increased at different phosphorus levels except for the decrease of Fe-P, the content of other inorganic phosphorus components increased, and the increase of organic materials reduced the proportion of soil Ca 10-P. Soil available P was positively correlated with Ca 2-P, Ca 8-P and Al-P( P<0.01). Path analysis showed that the direct contribution order of all forms of inorganic phosphorus to available phosphorus was Ca 2-P>Al-P>Ca 8-P>O-P>Ca 10-P>Fe-P, indicating that codomain Ca 2-P was the main available phosphorus source. Compared with P fertilizer alone, P fertilizer combined with organic materials significantly increased the yield and above-ground P content of winter wheat.

The accurate segmentation of wheat leaf disease and pest areas plays an important role in realizing the automation and intelligence of disease detection, and has important theoretical value and practical significance for improving the economic benefits of crops. In order to solve the problems of large workload and poor classification effect in the existing research, this study combines the active learning strategy of manual intervention labeling, and proposes a semi-supervised active learning method for wheat leaf disease and pest region segmentation. By extracting boundary features, color space features and texture features, the feature vector is composed, and the transductive support vector machine is used as the classifier. In each round of training, the k-means algorithm is used to randomly generate the samples to be labeled, and the human-computer interaction system is used to intervene the sample labels to improve the classification efficiency. This paper constructs a LM neural network model for classifying pests and diseases based on a new disease region segmentation method. In the image segmentation experiment, the proposed model is compared with the classical small sample SVM model and the semi-supervised TSVM model. The segmentation effect ranks first, which proves the superiority of the proposed segmentation strategy. In the experiment of pest identification, the model verification results showed that the recognition accuracy of LM neural network classification model using semi-supervised active learning method was 93.75%, which has good classification effect and can provide effective basis for subsequent pest control.

Optimization of extraction process and preliminary structural analysis of polysaccharide from Xiuzhen mushroom by response surface methodology

In order to study the optimal extraction process and preliminary structure of the polysaccharide from the Xiuzhen mushroom, the fresh mushroom fruiting body was used as the raw material, the polysaccharide content and iron reducing power were used as the indicators on the basis of the single factor experiment, and the extraction of the polysaccharide was optimized by response surface methodology. The structure was preliminarily analyzed by infrared spectroscopy and scanning electron microscope, and the thermal stability was analyzed. The results showed that the optimal water extraction process conditions for the polysaccharides of Xiuzhen mushroom were as follows: the extraction time was 3.2 h, the extraction times were twice, the extraction temperature was 90 ℃, and the ratio of liquid to material was 20 mL/g. The predicted value was 53.02 mg/g, the measured value was 53.98 mg/g, and the iron reducing power was 0.634. The measured results were in good agreement with the predicted values. The structure showed that the polysaccharide of Xiuzhen mushroom is a polysaccharide of pyranose configuration, with loose three-dimensional structure, smooth surface and hard texture. In this study, the extraction process of Xiuzhen mushroom polysaccharide was optimized, and the preliminary analysis confirmed that it was a polysaccharide substance, which provides a theoretical basis for the subsequent development and research of Xiuzhen mushroom polysaccharide functional substances.

Physiological responses of transgenic Arabidopsis thaliana with OsARP gene to salt stress

Salt stress has the effect of affecting the synthesis, metabolism, growth and development of plants in vivo. In order to investigate the effect of Auxin response protein ( OsARP) gene on salt stress in rice, four concentrations of NaCl (0, 50, 100, 200 mmol/L) were used to treat Arabidopsis thaliana with OsARP gene and wild-type Arabidopsis thaliana, and their agronomic traits and physiological indexes were measured. The results showed that the dry weight of transgenic Arabidopsis thaliana were 5 times higher than that of wild type, and fresh weight were 2 times higher than that of wild type under 200 mmol/L NaCl treatment. The root length was 93% higher than that of the wild type, and the plant height was twice higher than that of the wild type. The malondialdehyde (MDA) content was 25% higher than that of the wild type, and the soluble sugar content was 4.5% lower than that of the wild type. Under 100 mmol/L NaCl treatment, the chlorophyll content of transgenic Arabidopsis thaliana was 26% higher than that of wild type, and the activity of peroxidase (POD) was 17.3% higher than that of wild type, and the activity of catalase (CAT) was 7 times higher than that of wild type, and the activity of superoxide dismutase (SOD) was 15.6% lower than that of wild type. The results showed that the OsARP gene improved the salt tolerance of Arabidopsis thaliana, which laid a foundation for the study of the functional mechanism of OsARP gene.
