Effects of Ag ions and hormones on callus induction and differentiation of Chenopodium quinoa

Abstract: In this study, the effects of different concentrations of NAA hormone and different concentrations of sucrose on the callus of Chenopodium quinoa No.2 under different concentrations of Ag ions were studied based on MS medium. The results showed that in the medium MS+30 g/L sucrose +20 μmol/L Ag+ +0.8 mg/L NAA, C. quinoa No.2 seeds had the highest callus induction rate, reaching 100%. In medium MS+40 g/L sucrose + 20μmol/L Ag+ +0.4 mg/L NAA, C. quinoa No.2 seed had the lowest callus induction rate, only 13.3%. In medium MS+30 g/L sucrose + 20 μmol/L Ag+ +1.5 mg/L 6-BA, the differentiation rate of C. quinoa No.2 callus was the highest, which could reach 100%. In medium MS+20 g/L sucrose + 20 μmol/L Ag++2.0 mg/L 6-BA, C. quinoa No.2 callus had the lowest differentiation rate, only 20.0%. In this study, the optimal induction medium and optimal differentiation medium formula of C. quinoa No.2 callus were obtained, which will lay the foundation for the establishment of quinoa regeneration system and transformation system, and provide scientific basis for the preservation of quinoa germplasm resources, the cultivation of new varieties and factory breeding.
