The Physiological and Biochemical Characteristics Changes of Loose Salt-tolerant Callus of Alfalfa under Salt Stress

Abstract: In order to understand the physiological and biochemical changes of alfalfa salt resistant mutant, the content of soluble sugar, malondialdehyde, soluble protein, free proline and the cell membrane permeability, ionic concentration and activity of SOD & POD enzyme were studied with loose salt-tolerant and ordinary callus of alfalfa and under 2% salt stress. The results showed that the soluble sugar, POD enzyme activity, free proline and K+/Na+ of loose salt-tolerant callus was respectively 0.757, 0.266, 6.525 and 0.700 times higher than control group. However, the SOD enzyme activity, malondialdehyde, cell membrane permeability, and Na+ & K+ was 1.807, 0.878, 0.553, 2.903 and 1.296 times lower than control group. In conclusion, Loose salt-tolerant callus increased in soluble sugar, POD activity, free proline and the ratio of K+/Na+ under salt stress, while decreased in malondialdehyde cell membrane permeability and Na+, furthermore there was no significant change in soluble protein. In short , alfalfa mutations salt-tolerant callus have more resistance.
