Effects of Tissue Culture Conditions on Polysaccharide Contents of Dendrobium Draconis

Abstract: By tissue culture techniques, the various culture conditions (different concentrations of 6-BA, NAA, sucrose content and dark time) were adjusted to measure the weight and the content of polysaccharide from Dendrobium draconis at different culture state in this experiment in order to obtain the optimum culture conditions in which more polysaccharide contents of Dendrobium draconis were achieved. The results show that the optimum growth medium for Dendrobium draconis is 1/2 MS, 1.0 mg·L-1 6-BA, 0.3 mg·L-1 NAA, 6.5 g·L-1 agar and 25 g·L-1 sucrose. The highest polysaccharide content of Dendrobium draconis is the one which is continued to be cultured 7 days after 36 days of dark treatment.
