响应面法优化藜麦多糖提取工艺条件

Study on extraction process of quinoa polysaccharide by response surface methodogy

摘要: 本文分别采用水提取法、纤维素酶提取法、超声提取法3种方法,以藜麦多糖的提取率为考察指标,对红、白、黑3种颜色藜麦进行多糖提取。通过分析可知,采用纤维素酶提取法对3种颜色藜麦中的多糖进行提取的提取率均为最高,其中,白色藜麦多糖提取率最高,为6.8%,其次是红藜麦和黑藜麦,提取率分别为6.4%和5.9%;在此基础上,以白藜麦作为提取原料,采用单因素和Box-Behnken试验对提取料液比、加酶量、提取温度、提取时间进行优化,确定白藜麦多糖最优提取工艺条件为:料液比1∶32 g/mL,加酶量2 000 U/g,提取温度65 ℃,浸提时间90 min,在此条件下藜麦多糖提取率为8.0%。

Abstract: In this study, quinoa polysaccharide was extracted by water extraction method, cellulase extraction method and ultrasonic extraction method using the red color quinoa, white color quinoa and black color quinoa as material with the extraction rate of quinoa polysaccharide as an inspection index. The results showed that the highest extraction rate of polysaccharide was 6.8% with white quinoa through the enzyme extraction method, the red quinoa polysaccharide and black quinoa polysaccharide extraction rate were 6.4% and 5.9%, respectively. Furthermore, the single factor and Box-Behnken experiments were employed to optimize the cellulase extraction process for highest yield of white quinoa polysaccharides. The results suggested that the optimal extraction conditions were material-liquid ratio 1∶32 g/mL, the addition of cellulase 2 000 U/g, extraction temperature 65 ℃, and extraction time 90 min. Under the experiment conditions, the yield of polysaccharide was 8.0%.
