菌株ZYRH-19的筛选鉴定及其合成韦兰胶的特性研究

Screening, identification of strain ZYRH-19 and characteristics of its product welan gum

摘要: 从土壤中分离得到一株产胞外多糖的菌株ZYRH-19,从形态学、生理生化和16S rDNA基因序列的角度对其进行分类鉴定,并对其发酵合成的胞外多糖进行分析鉴别。结果表明:ZYRH-19为鞘氨醇单胞菌属菌株,与Sphingomonas sp. ATCC 31555同源性为99.93%,其合成的胞外多糖为韦兰胶,重均分子量为5.3×106 u;常温下韦兰胶溶液具有良好的稀释剪切性,在6 r/min剪切率下黏度能达到1 444.1 mPa·s,凝胶强度为22.6 g,在160 ℃下具有良好的抗温性能。研究表明,菌株ZYRH-19合成的韦兰胶性能更好,为韦兰胶的工业生产提供了新的菌种资源。

Abstract: In this study, a strain ZYRH-19 producing extracellular polysaccharide was isolated from soil. It was classified and identified from the perspectives of morphology, physiology, biochemistry, and 16S rDNA gene sequence, and the extracellular polysaccharide synthesized by strain ZYRH-19 was analyzed and identified. The results showed that ZYRH-19 was a strain of Sphingomonas sp., and its homology with Sphingomonas sp. ATCC 31555 was 99.93%. The extracellular polysaccharide synthesized by strain ZYRH-19 was welan gum with weight average molecular weight of 5.3×106 u. The welan gum solution synthesized by ZYRH-19 had good dilution shear property at room temperature. The viscosity of the product solution could reach 1 444.1 mPa·s at the 6 r/min shear rate, and its gel strength was 22.6 g. It had good temperature resistance at 160 ℃. The results showed that the performance of welan gum synthesized by strain ZYRH-19 is very good, which provides a new strain resource for the industrial production of welan gum.
