冰菜多酚的提取优化及抗氧化和抑菌活性研究

Study on extraction optimization of polyphenols from Mesembryanthemum crystallinum and its antioxidant and bacteriostatic activities

摘要: 以冰菜为材料,优化冰菜中多酚的提取方法,并测定多酚提取物的抗氧化能力和抑菌活性。结果表明,冰菜多酚的最优提取条件为:乙醇体积分数为60%,料液比1∶10(g: mL),提取温度80 ℃,提取时间120 min,此条件下多酚提取率为1.42%。冰菜多酚提取物对1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟基自由基的清除率随多酚质量浓度的增加而增加,当多酚提取物质量浓度为0.10 mg/mL时,对DPPH自由基和羟基自由基的清除率最大,分别为64.19%和88.10%。另外,冰菜多酚提取物对大肠杆菌的抑制作用明显,当提取物质量浓度为3 mg/mL时,抑菌圈直径为12 mm。

Abstract: In this study, the extraction method of polyphenols from Mesembryanthemum crystallinum was optimized, and the antioxidant capacity and antibacterial activity of polyphenols from M. crystallinum were determined. The results showed that the optimal extraction conditions were as follows: ethanol concentration was 60%, solid-liquid ratio was 1∶10(g: mL), extraction temperature was 80 ℃, extraction time was 120 min, under which the extraction rate of polyphenols was 1.42%. The scavenging rates of polyphenols from M. crystallinum to 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)and hydroxyl radicals increased with the increase of polyphenols concentration. When the concentration of polyphenol extract was 0.10 mg/mL, the scavenging rates of DPPH and hydroxyl radicals were 64.19% and 88.10%, respectively. The inhibitory effect of polyphenol extract on Escherichia coli was obvious. When the extract concentration was 3 mg/mL, the diameter of inhibition zone was 12 mm.
