当归多糖除蛋白质前后抗氧化活性比较

Compare of Antioxidant Activity of Angelica sinensis Polysaccharide before and after Protein Removing

摘要: 本试验比较当归多糖除蛋白质前后的体外抗氧化活性。采用水提醇沉法制备当归粗多糖(CASP),Sevag法除蛋白后得当归多糖(ASP)。用铁氰化钾还原法测定还原力、Fenton法测定羟基自由基清除能力,DPPH法测定自由基清除能力,邻苯三酚自氧化法测定超氧阴离子自由基清除能力。结果表明,CASP和ASP均具有显著的还原力和自由基清除能力(P<0.05);同浓度下,CASP的还原能力和自由基清除能力显著高于ASP(P<0.05)。但CASP和ASP的还原能力和自由基清除能力显著低于维生素C(P<0.05)。结果提示,CASP和ASP均具有抗氧化能力,CASP的抗氧化能力高于ASP。

Abstract: :This paper compared the antioxidant activity of Angelica sinensis polysaccharide(ASP)before and after protein removing. Crude A.sinensis polysaccharide(CASP)was extracted with hot water and precipitated with ethanol. The ASP was obtained by removing protein from CASP by Sevag method. Ferricyanide reduction method, fenton method, DPPH method, and adjacent benzene three phenol autoxidation method were adopted to assay the reduction ability and free radical-scavenging activity in vitro. The results showed that CASP and ASP exhibited significant reduction ability and free radical-scavenging activity(P<0.05). Within the same concentration, the reduction ability and free radical-scavenging activity of CASP were higher than those of ASP(P<0.05). However, the reduction ability and free radical-scavenging activity of CASP and ASP were feebler than those of ascorbic acid(P<0.05). These results indicate that CASP and ASP exhibited antioxidant ability, the antioxidant ability of CASP was stronger than that of ASP.
