Effects of stem section, sugar-free medium and environment on rooting of Prunus cerasifera Ehrh. plantlet in vitro

Abstract: In order to improve the in vitro rooting technology of Prunus cerasifera Ehrh. plantlet, the effects of different length of stem section, sugar-free medium and field environment on the rooting of Prunus cerasifera Ehrh. plantlet were studied. The results showed that the rooting rate of stem section inoculated in 15-20 mm length of Prunus cerasifera Ehrh. plantlet in vitro was 86.7%, significantly higher than that of stem section inoculated in 15-20 mm length. With the rooting rate of 90.0%, the root length of the plantlet in soil supporting medium without sucrose was significantly higher than that of the plantlet in medium containing sucrose. From the first ten days of September to the end of October, the rooting rate of Prunus cerasifera Ehrh. Plantlet cultured in the field environment was 83.3%, which was not significantly different from that of the plantlet cultured in the culture room. The results will provide the basis for improving the rooting culture technology and reducing the cost.
