观赏塔杨组织培养工厂化育苗

Technology of Tissue Culture and Factory Production for Ornamental Tower Poplar

摘要: 为快速繁殖塔杨,研究了组培快繁、试管苗入地、瓶外扦插和露地移栽等技术,总结出了塔杨组培工厂化育苗流程和技术要点。结果显示:带有顶芽的嫩梢接种到分化培养基MS+0.5 mg/L 6-BA+0.5 mg/L IAA上的芽增殖数较高;IAA和NAA均能使其芽100%生根,但根的质量差异较大;蛭石的移栽成活率高达92%,草炭仅30%,蛭石保湿培养1个月后移入混合基质营养钵可形成密集根系;100 mg/L的IAA或生根粉处理(速沾)嫩梢的瓶外扦插可使生根率和露地移栽成活率达80%以上。

Abstract: In order to propagate tower poplar (Populusⅹcanense) rapidly, the technologies of tissue culture, tube culture seedlings transplanting, cutting and cultivation were researched by hormonal combination, culture material combination, seedling hardening, seedling cultivation in the nutritive cups etc., and the factory breeding process for the tissue culture of Tower Poplar and the main technologies were summarized. The results show that more and better buds were gotten by young tip stem culture in the bud differentiation substrate of MS+0.5 mg/L 6-BA+0.5 mg/L IAA. The roots were produced in all plants by all concentrations of IAA and NAA in the test. But, the root quality and quantity were quite different. IAA treatment induced more roots compared with NAA. More and better roots were produced by the culture in the root induction substrate of 1/2 MS+0.25 mg/L IAA. The transplanting survival rate of seedlings was 92% on vermiculite, but only 30% on peat. The dense roots were grown in nutrition pots after seedlings had been cultivated in vermiculite for one month. The cutting propagation and cultivation management was relative to the selection of cutting, the hormone of rooting, and the culture materials and so on.
