Determination of Sulfonamides Residue in Chicken by High Performance Liquid Chromatography

Abstract: High performance liquid chromatography(HPLC)method was developed for the determination of sulfonamides residue in chicken, the chicken samples extracted with acetonitrile after homogenization, extracts were purified by Sep-Pak alumina column, determined by HPLC and ultraviolet UV detector, with acetonitrile methanol-water-acetic acid(2∶2∶9∶0.2, v/v)as mobile phase at a flow rate of 1.0 mL/min. The detection wavelength was 270 nm.The results showed that the peak area and concentration of the drug in the range of 0.1 to 5.0 mu g/mL, the correlation coefficient was greater than 0.999 6, the detection limit was 0.05 μg/kg, and the limit of quantification was 0.5 μg/kg. Among the 0.05, 0.20 and 0.50 μg/kg concentrations, the recovery was in the range of 60%~100%(n=7), and the relative standard deviation of the relative standard deviation was less than 15%. Compared with the traditional method, this method has a great improvement in detection speed and quantity.
