Isolation and Identification of Endophytic Fungi in Malus sieversii

Abstract: The endophytic fungi was isolated from M. sieversii stem using tissue separation method to obtain pure strains endophytes, the endophytic fungi species was identified using the molecular biology method, and the fungi ITS sequence was amplified and the comparation and analyses were made. The results show that 11 strains of endogenous fungi were identified from M. sieversii, which belonged to 3 species of endophytes in two genera(Alternaria sp. and Fusarium sp.), and there are same kind of endophytes among jujube, palm trees, M. sieversii and sugar cane. Pure strains can be obtained in short time initiated with MS medium and separated by PDA method. This method is simpler and more effective compared with multiple purified isolation method of PDA. These can provide experimental data for inhibiting the growth of endophytes generated from M. sieversii by the tissue culture.
