Establishment of a LAMP assay for detection of Enterocytozoon hepatopenaei

Abstract: In order to develop a rapid method for detecting the Enterocytozoon hepatopenaei (EHP), a loop-mediated isothermal amplification (LAMP) assay was established for targeting EHP-SSU gene with six specific primers following the optimization of the reaction. The results showed that the lowest limit detection of EHP-LAMP assay was 3.71 copies/μL, then its specificity was verified, and no cross-reactions was found for other shrimp pathogenic viruses including WSSV, IHHNV and CMNV. Moreover, 30 samples of shrimp with typical EHP infection symptoms were detected with the LAMP and PCR method, respectively. The results showed that the positive rates of LAMP and PCR were both 100%. In conclusion, the EHP-LAMP method is suitable for EHP early diagnosis with the advantages of high sensitivity, specificity and accuracy.
