Abstract: In this experiment, a single factor and orthogonal test methods were used to study the optimal culture conditions of a wild Trametes cubensis hyphae. The single factor test results showed that the optimal carbon source and nitrogen source for the mycelium of Trametes cubensis were glucose and yeast extract powder, and the optimal addition amount of carbon source and nitrogen source were 5 g/L, 3 g/L. The optimum culture temperature was 35 ℃, and the initial pH of the medium had no significant effect on the growth of the hyphae of Trametes Cubensis. Orthogonal test results showed that the order of the effects on the growth of the mycelium of Trametes Cubensis was: carbon source>temperature>nitrogen source. The results of orthogonal test showed that the optimum medium was glucose 15 g/L, yeast extract 3 g/L, magnesium sulfate 0.5 g/L, monopotassium phosphate 1 g/L, VB1 0.01 g/L, Agar 18 g/L, natural pH, culture temperature 36 ℃.
