谷氨酰胺在体内和体外试验中对鲤免疫力影响的研究

Effects of Glutamine on Immunity of Carp in vivo and in vitro Experiments

摘要: 以鲤的中肾白细胞作为研究对象,研究了谷氨酰胺对鲤免疫细胞免疫力的影响。在培养基中分别添加浓度为0、0.5、1.0、1.5、2.0 mmol/L的谷氨酰胺,在细胞培养2、6、12、24 h后,分别测定白细胞的呼吸爆发活力。结果发现,添加谷氨酰胺后,鲤肾脏白细胞的呼吸爆发活力得到了明显提高(P<0.05),且随着培养时间的延长,呼吸爆发活力呈现先上升后下降的趋势,最高值出现在培养12 h后的2.0 mmol/L谷氨酰胺组。在鲤肾脏白细胞培养12、24 h后,测定鲤肾脏白细胞的吞噬活力,发现白细胞吞噬活力变化趋势和呼吸爆发活力相似,其最高值出现在1.5 mmol/L谷氨酰胺细胞培养24 h后的添加组,鲤肾脏白细胞对迟钝爱德华氏菌杀菌率变化趋势与吞噬活力相符,添加谷氨酰胺后的各组杀菌率最高值发生在1.5 mmol/L谷氨酰胺组,与1.0、2.0 mmol/L谷氨酰胺组之间差异均不显著(P>0.05),但均显著高于对照组及0.5 mmol/L谷氨酰胺组(P<0.05)。将平均体重37 g的鲤鱼分为5组,每组30条,体内注射0、50、100、150、200 mg/kg鱼体重的谷氨酰胺溶液,商业饲料投喂2周后分离中肾获取鲤肾脏白细胞,发现白细胞的呼吸爆发活力和增殖活力随谷氨酰胺浓度的增加而呈现先显著上升后趋于平稳的趋势(P<0.05),而吞噬活力则一直显著升高(P<0.05),最高值均出现在200 mg/kg谷氨酰胺注射组。综合结果表明,谷氨酰胺是鲤有效的免疫增强剂。

Abstract: This experiments were conducted to evaluate the effects of glutamine on the immune responses of kidney leukocytes in common carp. Five different groups were formulated by supplementing 0,0.5,1.0,1.5,2.0 mmol/L glutamine into the basal culture medium. After the leukocytes were cultured 2,6,12,24 h,the respiratory burst activity of leukocytes were determined,respectively. The results showed that with glutamine supplemented into the culture medium,respiratory burst activity of leukocytes significantly improved and with prolongation of culture time and respiratory burst activity increased firstly(P<0.05),and then decreased. The peak occurred in 2.0 mmol/L glutamine group at 12 h. And then,leukocyte phagocytic activity was determined after cultured for 12 h and 24 h. The phagocytic activity change trend was similar with respiratory burst activity,the highest value occurred in 1.5 mmol/L glutamine group at 24 h. And no significant differences were found in 1.0,1.5, 2.0 mmol/L glutamine group,but the killing ability of leukocytesin these three groups were significantly higher than that in basal group and 0.5 mmol/L glutamine group(P<0.05). In vivo experiment,the average weight 37 g carp were divided into 5 groups. 0,50,100,150,200 mg/kg glutamine were injected to five different group. Then,fish was fed commercial diet for 2 weeks. Results demonstrated that with the increasing concentration of glutamine the kidney leukocyte respiratory burst and phagocytosis activity significantly increased firstly(P<0.05),and then leveled off,while the proliferation activity kept increased significantly(P<0.05). The results of this experiment indicate that glutamine is the effective immunomodulatory in carp leukocytes.
