锦带花组培快繁体系的建立

Establishment of tissue culture and rapid propagation system of Weigela florida

摘要: 以锦带花带腋芽茎段为外植体,探究最佳外植体消毒方法、最适初代培养基、最适增殖培养基,建立锦带花组培快繁体系。结果表明:外植体最佳消毒方法为2%次氯酸钠溶液消毒10 min后,接种至添加125 mg/L羧苄青霉素的MS培养基,外植体污染率仅1.67%,萌发率98.3%;初代培养最适基本培养基为WPM,组培苗平均株高可达5.58 cm、茎粗1.7 cm;最适增殖培养基为WPM+1.0 mg/L BA+0.5 mg/L IAA,增殖系数8.0,增殖芽高度1.7 cm,生长势强且未出现玻璃化现象。本研究可为锦带花工厂化生产和繁育提供有效的技术支撑。

Abstract: The stem segment of axillary bud was used as explants to explore the best method of explants disinfection, the best medium for primary generation and the best medium for proliferation, and to establish the tissue culture and rapid propagation system of Weigela florida. The results showed that the best disinfection method for explants was 2% sodium hypochlorite solution for 10 min, and then inoculated into MS medium supplemented with 125 mg/L benzylpenicillin. The contamination rate of explants was only 1.67%, and germination rate was 98.3%. The optimal medium for primary culture was WPM. The average plant height and stem diameter of tissue culture seedlings were 5.58 cm and 1.7 cm respectively. The optimal medium for proliferation was WPM+1.0 mg/L BA+0.5 mg/L IAA, the proliferation coefficient was 8.0, the height of bud was 1.7 cm, and the growth potential was strong without vitrification. This study can provide effective technical support for the factory production and breeding of Weigela florida.
