犬源伪中间型葡萄球菌的分离鉴定及药敏特性

Isolation, identification and drug resistance of canine Staphylococcus pseudointermediate

摘要: 天津某宠物医院接诊的一例患脓皮病的11岁金毛犬,采集其眼部、鼻部和皮肤分泌物,进行细菌分离培养。分离菌株TJ-1经培养特性、染色镜检、生化试验、血浆凝固酶试验、药敏试验及16S rDNA 序列分析,结果表明,TJ-1能分解果糖、乳糖、蕈糖、蔗糖、甘露糖、麦芽糖等,甘露醇和硝酸盐还原反应等为阴性,血浆凝固酶试验阳性;进一步采用PCR方法扩增16S rDNA序列,测序获得长度约为1 500 bp,构建系统进化树分析显示分离菌与Staphylococcus pseudintermedius聚为一支,最终确定TJ-1菌株为伪中间型葡萄球菌(S. pseudintermedius)。药敏试验的结果显示,TJ-1菌株对苯唑西林、头孢他啶、头孢哌酮、头孢氨苄、多西环素、头孢唑啉、米诺环素、头孢呋辛等药物敏感,对新霉素、红霉素、卡那霉素、四环素、庆大霉素、青霉素以及氨苄西林等耐药。

Abstract: An 11-year-old golden-haired dog with pyoderma was admitted to a pet hospital in Tianjin. The secretions of eyes, nose and skin were collected for bacterial isolation and culture. The results of culture characteristics, staining microscopy, biochemical test, plasma coagulase test, drug susceptibility test and 16S rDNA sequence analysis showed that the isolate TJ-1 decomposed fructose, lactose, mushroom sugar, sucrose, mannose, maltose, mannitol and nitrate reduction were negative, while plasma coagulase test was positive. The 16S rDNA sequence was amplified by PCR and sequenced. The phylogenetic tree analysis showed that the isolate was clustered with Staphylococcus pseudintermedius and TJ-1 strain was identified as S. pseudintermedius. Drug susceptibility test showed that the bacteria were susceptible to oxacillin, ceftazidime, cefoperazone, cefalexin, doxycycline, cefazolin, minocycline, cefradine, cefuroxime and amikacin, but resistant to erythromycin, penicillin, ampicillin and carboxybenicillin.
