Tissue culture and rapid propagation of Vitis vinifera ‘Muscat Hamburg’ cultured in soil supporting medium

Abstract: To get the technique of tissue culture and rapid propagation of Vitis vinifera ‘Muscat Hamburg’, the annual stem sections of ‘Muscat Hamburg’ were respectively picked up in late June and late September, sterilized with sodium hypochlorite and cultured in vitro. The sterile shoots proliferated in medium containing different concentrations of IBA, rooted in the soil-supported medium, and the transplanting with soil lump was carried out. The results showed that picking the stem sections on 27 June and soaking in 2% sodium hypochlorite for 10~15 minutes was better. In hormone-free GS medium, the propagation coefficient of test-tube plantlets could reach 3.6 but in the medium of 0.1~0.4 mg/L of IBA had no significant effect on proliferation. Two-node stem sections in the soil-supporting medium obtained the rooting rate of 84.4% which had no significant difference compared with test-tube plantlets cultured in agar medium, and the test-tube seedlings in soil-supporting medium had root hairs. The survival rate of test-tube seedlings cultured in soil supporting medium and transplanted with soil lump was 75.0% , which was significantly higher than that in agar medium. The results will provide a basis for the establishment of tissue culture and rapid propagation technology system of ‘Muscat Hamburg’.
