Optimization of determination condition of DHA-EE and EPA-EE in eggs by high performance liquid chromatography(HPLC)

Abstract: To detect the content of ethyl eicosapentaenoate(EPA-EE)and ethyl docosahexaenoate(DHA-EE)in derived egg samples by high performance liquid chromatography(HPLC), the super C18 chromatographic column was used to investigate the effects of proportion of organic phase, flow rate and column temperature on separation of EPA-EE and DHA-EE. The optimal chromatographic conditions were determined: isocratic elution, mobile phase of methanol and water(95:5, V/V), flow rate of 1.0 mL/min, column temperature 30℃, detection wave length of 220 nm. Through this method, the concentration and peak area of EPA-EE and DHA-EE showed a good linear relationship, and the correlation coefficient R2 were 0.999 2 and 0.999 0 respectively. And then the separate peaks of EPA-EE and DHA-EE in derived egg samples were detected. This method is rapid, simple and reproducible, and it provides ideas for the rapid detection of polyunsaturated fatty acids in eggs.
