Effects of 5-aza-dC and TSA on Development of Embryos Cloned from Different Donor Cells

Abstract: :To investigate the effects of 5-aza-dC and TSA on the development of embryos cloned from different donor cells, the fetal fibroblast cells, adult skin fibroblast cells, and cumulus cells were used as donor cells for nuclear transfer, the donor cells and cloned embryos were treated with TSA(50 nM, 12 h) and 5-aza-dC(20 nM, 72 h), and the effects of these reagents on the development of cloned embryos were investigated. The results show that 5-aza-dC combined with TSA could significantly promoted the development potential of cloned embryos (P<0.01), and the beneficial effects of TSA and 5-aza-dC on cloned embryos development were not of specific characteristics for the donor cells.
