运输期间维生素浸泡对红鳍东方鲀(Takifugu rubripes)生理生化指标的影响

Effects of vitamin bathing on physiological and biochemical parameters in pufferfish, Takifugu rubripes, during transport

摘要: 运输操作会对鱼类造成应激胁迫,口服B族维生素、维生素C有助于鱼类缓解应激胁迫,但用上述维生素药浴是否有助于缓解鱼类的应激反应,尚未见报道。本试验在体重为(23.8±3.8)g的红鳍东方鲀(Takifugu rubripes)运输期间(持续30 min),用维生素B2(10 mg/L)、维生素C(3.4 g/L)药浴,对照组在运输期间不加维生素。在运输结束后第一、三、五和七天取血液和组织,检测生理生化指标。结果表明,与维生素组相比,对照组白细胞总数、淋巴细胞总数、中间细胞总数、中性粒细胞总数、红细胞压积和血小板总数显著升高(P<0.05),肝胰脏超氧化物歧化酶(SOD)、溶菌酶(LZM)、天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、乳酸(LD)、过氧化氢酶(CAT)、丙二醛(MDA)、还原型谷胱甘肽(GSH)显著升高(P<0.05),脾SODCAT显著升高(P<0.05);对照组肌肉总抗氧化能力(T-AOC)、MDA显著升高(P<0.05),维生素组差异不显著;试验组肌肉LZMLDH变化显著(P<0.05),且对照组波动更大。结论在30 min的运输过程中用上述维生素浸泡,有助于维持红鳍东方鲀生理生化指标的相对稳定,缓解运输对其造成的胁迫。

Abstract: Transport manipulation can cause stress on fish, resulting in surface damage, deterioration of water quality, endocrine disorders, decreased immunity and even death. Vitamins as essential nutrients for the growth of aquatic organisms, play a role in anti-stress and immunity. Oral vitamins(mainly vitamin B, vitamin C)are important for aquatic animals, although oral administration of riboflavin and ascorbic acid contribute to relieve stress, whether these vitamins contribute to relieving fish stress via bathing has not been reported. In this test, riboflavin(10 mg/L)and ascorbic acid phosphate(3.4 g/L)were dissolved in sea water during Takifugu rubripes’s(23.8±3.8)g transport(lasting 30 minutes). In control group, no vitamin was added in sea water during transport. Physiological and biochemical parameters in blood and tissues were assayed at 1st, 3rd, 5th, 7th day post-transport. The results showed that the total number of white blood cells, lymphocytes, intermediate cells, neutrophils, erythrocytes and platelets were significantly higher in control group than those in vitamin group(P<0.05); Hepatopancreas superoxide dismutase(SOD), lysozyme(LZM), aspartate aminotransferase(AST), alanine aminotransferase(ALT), lactic acid(LD), catalase(CAT), malonaldehyde(MDA), glutathione(GSH), spleen SOD, CAT, and muscle total antioxidant capacity(T-AOC), MDA in control group increased significantly(P<0.05), and no significant difference was observed in vitamin group; Muscle LZM and LDH in both groups varied significantly(P<0.05). In addition, muscle LZM and LDH in control group varied greater than those in vitamin group. In general, the physiological and biochemical parameters changes in control group after transport were obviously greater than those in vitamin group. In conclusion, bathing with riboflavin and ascorbic acid phosphate during 30-minute transport contributed to relieving transport stress of pufferfish, Takifugu rubripes, which provides a reference for its practicle transport.
