白桦茸胞外多糖的抗氧化性研究

Study on the antioxidant activity of exopolysaccharides from Inonotus obliquus

摘要: 白桦茸(Inonotus obliquus)是一种含有多种生物活性物质的食用菌,其多糖成分具有良好的抗氧化活性。本研究从白桦茸液体发酵液中得到胞外粗多糖,采用苯酚-硫酸比色法测定其含量,并比较除蛋白前后胞外多糖的体外抗氧化能力。结果表明,白桦茸液体发酵液中粗多糖含量为1.28 mg/mL;采用Sevag法脱蛋白,脱蛋白次数为4时,蛋白去除率达51.5%,多糖损失率为25.4%;体外抗氧化试验显示,当质量浓度为0.8 mg/mL时,脱蛋白前白桦茸胞外多糖对羟自由基(·OH-)、超氧阴离子自由基(O2)和DPPH的清除率分别为33.1%、33.4%和32.1%,脱蛋白后对应清除率分别为29.9%、30.6%和25.6%,表明白桦茸胞外多糖具有一定的体外抗氧化活性。

Abstract: Inonotus obliquus is a kind of edible fungus containing a variety of bioactive substances, among which its polysaccharides exhibit notable antioxidant activity. In this paper, extracellular crude polysaccharide was obtained from the liquid fermentation broth of I. obliquus, and the antioxidant ability of extracellular polysaccharide from I. obliquus before and after deproteinization was detected. The content of crude polysaccharide in liquid fermentation broth of I. obliquus measured by phenol-sulfuric acid colorimetric method was 1.28 mg/mL. The deproteinization rate reached 51.5%, but the loss rate of polysaccharide was 25.4% with Sevag method after four rounds of deproteinization. In vitro antioxidant studies, the concentration was 0.8 mg/mL, the clearance rates of ·OH-, O2and DPPH were 33.1%, 33.4% and 32.1%, but after deproteinization the clearance rates for ·OH-, O2and DPPH were 29.9%, 30.6% and 25.6%, respectively. It is concluded that extracellular polysaccharide of I. obliquus has certain antioxidant ability in vitro.
