白藜芦醇对ZEN诱导的猪小肠上皮细胞损伤的作用效果研究

Effects of resveratrol on zearalenone induced injury of porcine intestinal epithelial cells

摘要: 试验旨在探究白藜芦醇对玉米赤霉烯酮(ZEN)诱导的猪小肠上皮细胞(IPEC-J2细胞)损伤的影响。试验分组为:C组(对照组)、Z组(ZEN浓度25 µg/mL)、ZR1组(25 µg/mL ZEN+10 µmol/L白藜芦醇)、ZR2组(25 µg/mL ZEN+20 µmol/L白藜芦醇)、ZR3组(25 µg/mL ZEN+30 µmol/L白藜芦醇);测定细胞活力、细胞乳酸脱氢酶(LDH)活性、抗氧化酶水平以及Caspase 3Caspase 9Bcl-2bax等凋亡相关基因mRNA表达水平。试验结果显示:Z组细胞活力、SOD活性T-AOC水平和Bcl-2基因mRNA表达水平极显著低于C组(P<0.01),ZR2组和ZR3组细胞活力、SOD活性、T-AOC水平和Bcl-2基因mRNA表达水平极显著或显著高于Z组和ZR1组(P<0.01或P<0.05);Z组细胞LDH活性、MDA水平、Capase-3Capase-9Bax基因mRNA表达水平极显著高于C组(P<0.01),ZR2组和ZR3组细胞LDH活性、MDA水平、Capase-3Capase-9Bax基因mRNA表达水平极显著或显著低于Z组和ZR1组(P<0.01或P<0.05),ZR1组细胞LDH活性和Bax基因mRNA表达水平极显著低于Z组(P<0.01)。结果表明,白藜芦醇能够缓解ZEN诱导的IPEC-J2细胞的细胞活力、LDH活性、抗氧化功能和凋亡相关基因mRNA表达水平的改变。

Abstract: The purpose of this study was to investigate the effect of resveratrol on the injury of porcine small intestinal epithelial cells(IPEC-J2 cells)induced by zearalenone(Zen). The experimental groups were: Group C(control group), group Z(25 µg/mL Zen), group ZR1(25 µg/mL Zen+10 µmol/L resveratrol), group ZR2(25 µg/mL Zen+20 µmol/L resveratrol), group ZR3(25 µg/mL Zen+30 µmol/L resveratrol). The cell viability, the activity of lactate dehydrogenase(LDH), the level of antioxidant enzymes and the mRNA expression of apoptosis related genes such as caspase 3, caspase 9, Bcl-2 and Bax were measured. The results showed that the cell viability, SOD activity, T-AOC level and Bcl-2 gene mRNA expression level in group Z were significantly lower than those in group C(P<0.01). The cell viability, SOD activity, T-AOC level and Bcl-2 gene mRNA expression level in group ZR2 and ZR3 were significantly higher than those in group Z or group ZR1(P<0.01 or P<0.05). The LDH activity, MDA level, Capase-3, Capase-9 and Bax gene mRNA expression levels in group Z were significantly higher than those in group C(P<0.01). The LDH activity, MDA level, Capase-3, Capase-9 and Bax gene mRNA expression levels in group ZR2 and ZR3 were lower or significantly lower than those in group Z or group ZR1(P<0.01 or P<0.05). The LDH activity and Bax gene mRNA expression levels in ZR1 group were lower than those in group Z(P<0.01). The results indicated that resveratrol could alleviate the changes of cell viability, LDH activity, antioxidant function and apoptosis related gene mRNA expression of IPEC-J2 cells induced by Zen.
