番泻叶致小鼠急性腹泻模型的建立

Establishment of acute diarrhea model in mice caused by Folium Sennae

摘要: 本研究旨在建立小鼠急性腹泻模型。取昆明小白鼠30只,随机分为3组,分别为模型组1、模型组2及对照组,每只分别灌胃1 g/mL番泻叶水煎液0.2 mL、0.3 mL和生理盐水0.2 mL。每天两次,连续5天。观察小鼠排便情况,并记录稀便率、稀便级、腹泻指数。试验第六天,每组随机选取小鼠5只采集血清,测定血糖和甘油三酯。采血后断颈处死,采集回肠和盲肠标本,制作苏木精-伊红(HE)染色病理切片。通过指标稀便率、稀便级、腹泻指数及体重分析,两种番泻叶浓度均可明显致小鼠急性腹泻(P<0.05),并对小鼠血糖水平无明显影响(P>0.05),但高浓度番泻叶使甘油三酯血液水平有明显降低(P<0.01)。饲喂番泻叶组小鼠的回肠盲肠黏膜均无明显坏死和炎症反应,但高浓度番泻叶会影响肠道黏膜下层间隙并使少量绒毛上皮细胞脱落。结果表明两种浓度番泻叶均可建立小鼠急性腹泻模型,但灌胃0.3 mL番泻叶组效果更明显。

Abstract: The experiment was conducted to set up the acute diarrhea model of the mice. 30 mice were randomly divided into 3 groups named as following: Model group 1, Model group 2 and control group. Each of the mice were all fed 1 g/mL basal diet, respectively plus 0.2 mL Folium Sennae, 0.3 mL Folium Sennae and 0.2 mL normal saline. Each of the three drug-delivery were processed twice every day. The duration was five days. The defecation of the mice was observed and the indexes of loose stool rate, degree of diarrhea, diarrhea index were tested. Blood of half of the mice in every group were collected through the eye socket at the sixth day after starting the experiment, which were used to measure blood sugar and triglycerides. All of the mice with blood collected were killed with their necks severed for collecting the samples of ileum and cecum which were used to make pathological sections of hematoxylin and eosin. The results showed that both levels of the two group intake Folium Sennae have developed the mice acute diarrhea model(P<0.05)and had no significant effect on blood sugar of the mice. The blood triglycerides of the mice intake higher dose Folium Sennae significantly decreased(P<0.01). Neither groups of taking Folium Sennae had obvious necrosis or inflammation of intestinal mucous. The result showed that the two doses of Folium Sennae can establish acute diarrhea model successfully, much better with higher dose group(0.3 mL).
