Effects of MicroRNA-139 on Myofiber of Denevated Muscle Atrophy in Mouse

Abstract: To investigate the effects of miR-139 on myofibers of denervated muscle atrophy in mouse, anamyotrophia mouse model of sciatic nerve resection was established. 10 nmol miR-139 inhibitors were injected into the denervated gastrocnemius. 14 days after injection, the expression of miR-139 was detected by qRT-PCR, the wet weight and the cross section diameter of the muscle fibers were compared, and the expression of myosin heavy chain(MHC)was examined by RT-PCR and Western blot analysis between the atrophying skeletal muscle and control. The results showed that the cross section diameter of the muscle fibers in treated group was significantly higher than that of control group. The results suggested that miR-139 may play an important role in muscle growth and differentiation in vivo.
