油用牡丹组培快繁研究

Study on tissue culture and rapid propagation of Paeonia ostii

摘要: 为建立油用牡丹组培快繁体系,本研究以油用牡丹腋芽为外植体,采用不同浓度的消毒剂进行表面消毒,获得无菌苗;将无菌苗分别接种到含有不同激素的MS培养基上,进行侧芽的增殖培养;将获得的再生苗接种到含有不同浓度的生长素培养基上进行生根培养。结果显示:采用15%的安替福民,处理5 min,获得的消毒效果最好;在MS+1.0 mg/L BA+0.5 mg/L IAA+5 mg/L AgNO3培养基上油用牡丹再生芽增殖效果最好;在MS+0.1 mg/L NAA+0.5 g/L AC培养基上再生苗生根效果最好。本试验结果可以为油用牡丹的组培研究奠定基础。

Abstract: In order to establish the rapid propagation system of Paeonia ostii tissue culture, this study used Paeonia ostii axillary buds as explants and used disinfectant of different concentrations for surface disinfection to obtain bacteria-free seedlings.The results showed that the best disinfection effect was obtained when 15% antihuman was used as disinfectant for 5min. In MS+1.0 mg/L BA+0.5 mg/L IAA+5 mg/L AgNO3 medium, the regeneration bud proliferation effect of Paeonia ostii was the best.The best rooting effect was observed in MS+0.1 mg/L NAA+0.5 g/L AC medium. The results of this experiment can lay a foundation for the study of tissue culture of Paeonia ostia.
