依赖SRP对大肠杆菌膜蛋白定位的研究

Localization of E.coli membrane proteins dependent on SRP

摘要: 大肠杆菌中,信号肽识别颗粒(signal recognition particle,SRP)识别并绑定核糖体正在翻译的信号肽,引导新生肽链到细胞膜上。根据靶向膜蛋白生物素酰化原理,本研究利用Western blot方法,对构建的HDB51菌株及野生型菌株MG1655进行大肠杆菌膜蛋白定位研究。对比不同菌株中是否含有SRP,结果发现,在缺失SRP的HDB51菌株中检测到依赖SRP的底物蛋白FtsQ 的定位,缺失SRP 明显影响了FtsQ的定位,SRP具有辅助膜蛋白定位的功能,从而为研究依赖SRP性大肠杆菌膜蛋白定位提供理论依据。

Abstract: In Escherichia coli, signal recognition particle(SRP)recognizes and binds signal peptides being translated by ribosome to guide the new peptide chain to the cell membrane. In this experiment, localization of E.coli membrance protein in HDB51 and MG1655 was studied by Western blot based on biotinylation for membrane proteins targeting. By comparing the presence or absence of SRP in different E.coli, the substrate protein FtsQ that depended on SRP in the strain E. coli HDB51 was localized. The location of FtsQ was significantly affected without SRP. This verifies that SRP helps membrane protein localization. It can provide references for the study of localizing E. coli membrane proteins dependent on SRP.
