革胡子鲶干扰素α基因对维氏气单胞菌感染的应答分析

Response of interferon α gene to Aeromonas veronii infection in Clarias gariepinus

摘要: 应用实时荧光定量PCR技术分析了革胡子鲶感染维氏气单胞菌前后其脾脏、肾脏、头肾、鳃、肠道、皮肤中干扰素α基因的表达谱。结果显示:该基因在健康(对照组)革胡子鲶的6种组织中均有表达,但以脾脏表达量最高,显著高于除肾脏和皮肤以外的其他组织;感染病原菌后3~96 h,该基因在上述6种组织中的时序表达模式相似,即:表达水平首先显著上调、之后回到对照组水平,但到达峰值的时间不同;此外,在感染病原菌后的相同时间点,该基因的表达水平有组织差异性。

Abstract: In this paper, real-time quantitative PCR technique was used to assess the expression profiles of interferon alpha(IFNα)gene in spleen, kidney, head kidney, gill, intestine and skin of Clarias gariepinus following an intraperitoneal injection of Aeromonas veroni. The results showed that the IFNα gene was expressed in all tissues of the healthy(control)individuals, and the highest expression was found in spleen, which was significantly higher than those in other tissues except kidney and skin. After A. veronii challenge during 3-96 h, the temporal expressions profiles of IFNα gene in the six tissues were similar, namely, transcripts were up-regulated firstly and then recovered back to the baseline, but the expression peaked at different time points. Furthermore, the expression levels of IFNα gene were different among the tissues at the same time points of post infection.
