Effects of Environments on in Vitro Rooting and Survival of Malus zumi Micro Shoots

摘要: 本文旨在探讨在露地和温室环境中珠美海棠微枝试管内生根的可能性。将珠美海棠试管苗茎段接种于土壤支撑的培养基中,分别置于露地、温室和恒温培养室进行生根培养,并对培养的试管苗进行气孔观察和移栽试验。结果表明,3种环境下培养的珠美海棠试管苗生根率无显著差异,温室和露地中的试管苗根长极显著高于恒温室的试管苗,温室和露地培养的试管苗叶片气孔的关闭功能明显强于恒温室的试管苗。在不经过炼苗直接移栽的条件下,露地和温室环境中培养的试管苗移栽成活率均达96.7%,显著高于恒温培养室的试管苗。在温室和露地环境中进行珠美海棠微枝的试管内生根培养具有可行性,值得进一步研究。

Abstract: This test was designed to investigate the possibility of Malus zumi micro shoots in vitro rooting in the environmental conditions of both field and green-house. The Malus zumi micro shoots were inoculated in the soil supporting medium and placed respectively in field, greenhouse and incubation chamber for rooting culture, and then the stomata of the plantlet cultured was observed and transplanting test was done. The results show that there were no significant differences in rooting rate among three environmental conditions. The root length of the plantlet cultured in greenhouse and field were significantly higher than that of the plantlet cultured in incubation chamber. The stomata closing function of the plantlets cultured in field and greenhouse were stronger than that of the plantlet cultured in incubation chamber. Being transplanted without hardening the plantlet cultured in field and greenhouse had survival rate of 96.7%, which was significantly higher than that of the plantlet in incubation chamber. In vitro rooting of Malus zumi micro shoots in greenhouse and field environment is feasible and worthy of further study.
