矮溲疏试管苗的土壤支撑生根培养和带坨移栽

Rooting in Soil Supporting Medium and Transplanting with Lump of Deutzia hybrida ‘Boule’ Plantlet in Vitro

摘要: 以土壤做培养基的支撑物,在培养基中分别加入不同浓度(0、5、10、15 g·L-1)的蔗糖和不同浓度(0,0.2、0.4、0.6、0.8 mg·L-1)的IBA,并进行矮溲疏试管苗茎段的生根、开瓶炼苗和帯坨移栽,以及开瓶炼苗期间试管苗的气孔开闭状况的研究。结果表明,在不添加蔗糖和IBA的无菌土壤中,矮溲疏试管苗的生根率可达100%,根长及植株地上部分的生长与添加蔗糖和IBA的处理无明显差异;在不覆膜、不喷雾、午后空气相对湿度接近40%的移栽条件下,帯坨移栽矮溲疏试管苗的成活率为100%,显著高于常规的苗木移栽方法(72.2%);开瓶炼苗14 d后,黑暗中土壤支撑培养的矮溲疏试管苗叶片气孔的半关闭率由未开瓶时的1.3%提高到98.7%,气孔相对开张度由未开瓶时的0.618降低到0.193。该结果为进一步完善矮溲疏组织培养快速繁殖技术体系提供了依据。

Abstract: Different concentrations (0, 5, 10, 15 g·L-1) of sucrose and different concentrations (0, 0.2, 0.4, 0.6, 0.8 mg·L-1)of IBA were respectively added to media supported by soil, and the stem section rooting of the Deutzia hybrida ‘Boule’ plantlet in Vitro, acclimatizing in opened culture bottle, transplanting with lump and the stoma closing state of the plantlet during acclimatized in opened culture bottles were studied. The results show as follows. In the sterile soil without sucrose and IBA added, the rooting rate of the Deutzia hybrida ‘Boule’ plantlet was 100%, and the root length and the stem and leaf growth of the plantlet were not significantly different from those of the plantlet in medium containing sucrose or IBA. Under the transplanting conditions without plastic film covering or spraying and the air humidity decreased to 40% in afternoon, the survival rate of the plantlet transplanted with lump was 100%, which was obvious higher than that of the plantlet transplanted with normal transplanting method (72.2%). After acclimatized in opened culture bottles for 14 days, the half closing rate of the stoma in leaf of the plantlet cultured in soil supporting medium in dark increased from 1.3% in no opened culture bottles to 98.7%, while the stoma relative opening degree decreased from 0.618 in no opened culture bottles to 0.193. The results provide the basis for improving the rapid micropropagation system of Deatzia hybrida ‘Boule’.
