Optimization of culture conditions for astaxanthin production by Haematococcus pluvialis with response surface methodology

Abstract: Haematococcus pluvialis is an important resource microalgae. The product astaxanthin has excellent antioxidant activity and is widely used in medicine and food. In order to study the high-yield astaxanthin of Haematococcus pluvialis, three stress conditions(5 000, 7 000, 9 000 lx), sodium nitrate concentrations(0, 0.13, 0.25 g/L)and chlorination were studied. The effects of sodium concentrations(0, 2.5, 5.0 g/L)on astaxanthin content of Haematococcus pluvialis were optimized by response surface methodology. The order of influence of the three factors on the production of astaxanthin content was: sodium chloride concentration>light intensity>sodium nitrate concentration. The optimal extraction conditions were: light intensity 9 000 lx, sodium nitrate concentration 0 g/L, sodium chloride concentration 2.5 g/L, under which the astaxanthin content could reach 6.8 mg/L, and it was similar to the predicted value. The results indicated that the model has high reliability.
